Objective: In order to compare the biological characteristics of human mesenchymal stem cells derived from bone marrow and placental decidua basalis in the three-dimensional culture system set with small intestinal submucosa and resistant to the hypoxia and serum deprivation and to provide theoretical rationales for selecting the tissue engineering seed cell.Methods: Mesenchymal stem cells(MSCs)were extracted from human bone marrow and human pacental decidua basalis tissues in a sterile manner and then amplifying cultured in vitro.The Surface marker Identification of BM-MSCs and PDB-MSCs is made By Flow Cytometry.At the same time,the differentiation potency of PDB-MSCs and BM-MSCs toward the adipogenic,osteogenic,and chondrogenic lineages were investigated.The MSCs(5 × 105 cells/cm2)were seeded on the sis and obverse the morphology of PDB MSCs and BM-MSCs on SIS.the cells-scaffold compounds were formed and then the cells-scaffold compounds were randomly divided into 4 groups: BM-MSCs(normoxia and complete medium,N/CM)? BM-MSCs(hypoxia and serum deprivation,H/SD)?PDB-MSCs(normoxia and complete medium,N/CM)?PDB-MSCs(hypoxia and serum deprivation,H/SD).Later,the growth curve of BM-MSCs and PDB-MSCs was drawn by a cell counting kit–8(CCK8)cell proliferation assay kit.The anti-apoptotic ability of the PDB-MSCs and BM-MSCs were achieved by an Annexin V/propidium iodide apoptosis assay kit.The Expression of VEGF were detected using Q-PCR assay;the Expression proteins of AKT?ERK1/2?Bcl2 were also detected using Western Blot assay.Results: MSCs derived from human bone marrow and human pacental decidua basalis tissues were successfully extracted and cultured in vitro.The two kinds of cells were positive for CD90,CD73 and CD105 and negative for CD34,CD11 b,CD19.What is more,BM-MSCs and PDB-MSCs were differentiated into osteoblasts,adipocytes and chondrocytes.The cck8 assay showed the PDB-MSCs in serum-free conditions under hypoxia grew faster than the BM-MSCs(P<0.05).the AV/PI assay showed the PDB-MSCs tolerance stronger than the BM-MSCs in serum-free conditions of hypoxia(P<0.05).After induced with hypoxia and serum deprivation,the two kinds of cells expressed the m RNA of VEGF.What is more,compared with BM-MSCs,PDB-MSCs expressed much more m RNA of VEGF(P<0.05).After induced with hypoxia and serum deprivation,the two kind of cells both expressed the protein of AKT?ERK1/2?Bcl2.What is more,compared with BM-MSCs,PDB-MSCs expressed much more specific protein of AKT?ERK1/2?Bcl2(P<0.05).Conclusion: SIS is an ideal bio-scaffold material for cardiac tissue engineering,it has good biocompatibility with BM-MSCs and PDB-MSCs.the PDB-MSCs tolerance stronger than the BM-MSCs in serum-free conditions of hypoxia.Thus,PDB-MSCs may be more fit to seed cells for myocardial tissue engineering.
