Malate dehydrogenase plays an important role in life activities,and it is involved in many physiological activity of cells.Through researching the gene differential expression between normal bud and abortion bud,Our research group(Jia jin et al.2009)speculated there may exsit some relationship between malate dehydrogenase gene(BrMDH)and floral bud abortion(FBA)in Chinese cabbage by comparing the gene differential expression between normal flower bud and FBA.However,the function what the malate dehydrogease play in Chinese cabbage growth is not clear.In this study,the full length of BrMDH was obtained from FBA of Chinese cabbage "97C16-3".The expression of BrMDH in different tissues of Chinese cabbage were analysed with the real-time quantitative PCR technology.Later the over-expression vector p2301-BrMDH and a RNA interference(RNAi)expression vector pRNAi-LIC-BrMDH were constructed and transformed into wild-type Arabidopsis by the method of Agrobacterium mediated.The transgenic plants were identificated and their phenotypes were observed.This study set a good foundation for us to study the function of BrMDH gene and research the mechanism of FBA.The main results were as follows:1?BrMDH is cloned from Chinese cabbage,its sequence had one exon with none intron.2?By analysing the expression of BrMDH in different tissues of Chinese cabbage with real-time quantitative PCR technology,we found that the gene express highly in floral bud which turning from normal to abortion and the apical meristem of inflorescence.The most important was that the expression of BrMDH showed a trend which increased firstly than reduced,as the normal bud tun to FBA.The abnormal express indicated that there was a close relationship between BrMDH and FBA.3?The over-expression vector p2301-BrMDH and RNAi expression vector pRNAi-LIC-BrMDH were constructed and transformed into wild-type Arabidopsis by the method of Agrobacterium-mediated,transgenic plants were obtained.4?By observing the transgenic plants,we found some transgenic arabidopsis showed FBA when the MDH expression was interfered,this mean that there was a some relationship between MDH and FBA.In addition,MDH was involved in the formation of lateral branch,rosette leaves colour and the formation of chlorophyll,plant height,pod length in Arabidopsis.
