sRNAs(Bacterial small RNAs),belongs to nocoding RNA family.It's length is between 40-500nt.sRNAs is found to participate many metabolism progress in bacterial resent years.With the fast development of High-throughput sequencing,more and more sRNAs of Xanthomonas campestris pathovar campestris(Xcc)strains 8004 are identified in our lab.This job is to study the fuction of sRNAs which we name sRNA-Xcc 19,sRNA-Xcc20,sRNA-Xcc21,sRNA-Xcc22,sRNA-Xcc23,sRNA-Xcc24.We construct of six sRNAs genes "over expression strains" and one "mutant deletion strain" and name them OE19,OE20,OE21,OE22,OE23,OE24,DM19.Phenotypes analysises show that,OE23 and OE24 have some changes compared with the wide type Xcc8004 and Xcc8004p(the control strain that induced an empty plasmid).OE23 displayes a significant reduction in the production of extracellular protease and repressed by high metal ions concentration of Zn2+,Cu2+,Cd2+,Ni2+ and MMX medium and SDS;The infection ability of OE23 to it's host is slightly decrease.At the same time,OE24 is repressed by MMX medium,it's growing ability on minimal medium is much more slower than that on the NYG medium.Most meaningful finding is that OE24 almost loose it's infection ability to it's host.While,other five strains and the mutant strains have no dramatical changes.More study should be done to get to know if the genes have any connection with metabolism and pathology including constructing the delection mutants and their complementary strains.
