ABSTRACT A large number of reservoirs distributed over China's major tributaries of rivers charge with people's normal production and living water around the Yellow River valley.The water quality safety is related to people's health and the quality of life and is closely linked with people's living.At present,the water pollution and the shortage of water resources are increasingly serious what is happening to China.The thesis studies the biological diversity and how microbial communities change with the change of the complete water period in the Yellow River(Xiaolangdi reservoir).That can give a basis to studying the reservoir ecosystem and the development of reservoir water biological treatment programs.This research takes Xiaolangdi reservoir in the Yellow River as the area to studying,setting up a total of nine sampling transects.In different water period(level water period,abundant water period and dry water period),we took samples of each section of Xiaolangdi reservoir and analyzed the water quality in Xiaolangdi reservoir and the microbial community diversity using the traditional water quality monitoring,the microbiological analysis method and PCR-DGGE.This article contains four parts and the conclusions are as follows:(1)Analysing the water quality paramenters from March to April in 2011 shows that the water quality of the Xiaolangdi reservoir are ? water quality for the ammonia nitrogen exceeds the standard.In other monthes,the quality is ? or ?.It is better.(2)The total number of bacteria changes obviously with water the phase,accounting for the total number of bacterias.The colony number of the flat and abundant water period is higher,but that of low water period is relatively lower.Besides dry season individual cross sections in 100 CFU/mL,others are in 100 mL above.That is the total number of bacteria severely exceed the standard of water.(3)According to the investigation of phytoplankton,Xiaolangdi reservoir's phytoplanktons are mainly green algaes and diatoms,in the dominant species composition.The dominant species are mainly algae,the needle bar straight chain,Navicula,C.cohnii,green algae,chlorella,empty cell.Each section of the dominant species also occupy certain advantages.The trend to the number of algaes' change is that the number in the mean water period is most,in the abundant water period secondary and in the dry season least.In all,it reveals tall at first and low behind.(4)By analyzing the biodiversity of the Xiaolangdi reservoir in PCR-DGGE,we obtained that the reservoir microbial is very multifarious and changes with the dry season.The biodiversity in the abundant water period is the most abundant and that of the mean water period is the worst.The trend assumes gradually increased.Dominant species are diverse,mainly including Malikia,Flavobacterium,Bosea,Deinocoaus etc.The Bosea and Flavobacterium appear with high frequency.(5)Compared to water quality characteristic parameters,the total number of bacterias,phytoplankton quantity and biodiversity,the total number of bacteria and the water temperature is,the more the number is.However,the other water quality parameters change unobviously.The number in the mean water period is the largest,related to the water body's high content of ammonia nitrogen from March to April.The more ammonia nitroger is,the more the number of algae is.The trends of phytoplankton and microorganism are just opposite with the complete water period.Those present that the more phytoplankton is,the less microbial is.
