It is very difficult to identify Culex molesuts and Culex quinquefasciatus by their morphology.The former belongs to autogenous mosquitoes and has the ability to develop a batch of eggs without a reliance on a blood meal.The latter is an anautogenous mosquito and must take a blood meal in order to supply nutrients for egg development,which is responsible for the transmission of vector-borne disease.There is a direct relationship between reproductive process and feeding habit.However,the molecular mechanism of the reason causing the different feeding habit is unkown.In this study,high-throughput sequencing Hiseq 2500 was employed to sequence the female midgut transcriptome of Cx.molesuts response to non-feeding and sugar-feeding,respectively.A total of 11.94 M and 17.43 M reads and 493 differential expression genes were obtained.The female midgut transcriptome of Cx.quinquefasciatus was sequenced following non-feeding,sugar-feeding and blood-feeding.A total of 13.24 M,11.45 M and 13.43 M reads were obtained.We also obtained 785 and 840 differential expression genes following sugar-feeding and blood-feeding,respectively.Based on the comparison analysis of the two female midgut transcriptome following non-feeding and sugar-feeding,we obtained 1640 and 1558 differential expression genes,respectively.The differential expression genes were annotated with Gene ontology(GO),Kyoto Encyclopedia of Genes and Genome(KEGG)database which involved in digestion,reproduce,detoxification,immunization.Cx.molesuts has a greater abundance transcription of sugar digestion genes and serine protease genes compared with Cx.quinquefasicauts.There was a huge difference in vitellogenesis between two species.Cx.molesuts started the synthesis of vitellogenin,trpsin and chymotrypsin as soon as eclsoion.Cx.quinquefasciatus transcribed vitellogenin genes only induced by blood meal but transcript of apolipoprotein has a greater abundance after eclosion.The results reveal transcription level of important genes in different feeding conditions.We attempt to explore the similarities and differences of metabolism and embryonic development between anautogeny and autogeny.The results obtained were of great importance for the functional genes research and development of novel insecticides.
