| The auditory system is one of the important sources of information for human and animals.Inner ear hair cells in the auditory system(HC)bear the function of transforming mechanical signal into electrical signal,hair cells are necessary for the normal function of auditory.The top of the hair cells are formed by the actin monomers(G-actin)polymerization ladder shaped arrangement of the three rows of stereocilia,the morphological and structural integrity of the stereocilia is essential to the normal auditory transduction.The protein FCHSD2 is the homologous protein of NWK in Drosophila,which is one member of F-BAR protein family.F-BAR proteins contains a BAR domain and involve in the regulation of actin polymerization,closely related with the reshaping of the cytoskeleton.The lack of NWK would cause nerve cells around the neuromuscular junction excessive growth and sustained release neurotransmitters,ultimately killing Drosophila.The study of FCHSD2 rarely reported,The preliminary study of our research group shows that FCHSD2 can directly combine WASP/N-WASP and promote Factin polymerization in vitro.The immunohistochemical results showed that FCHSD2 was distributed along the auditory stereocilia.Our initial guess FCHSD2 may play a role in the auditory system in mice.in order to further study whether FCHSD2 participated in the process of developing and maintaining of the stereocilia.in turn affects the function of the auditory system in mice.We use the homologous recombination and CRISPR/Cas9 technology to construct Fchsd2 gene knockout mice.Hearing detection of Fchsd2 gene knockout mice found that the deficiency of the protein FCHSD2 leaded to age-related hearing loss in mice,and Fchsd2 knockout mice appeared the lack of inner ear hair cells and the disorganization of stereocilia during the late development.But the exact mechanism is unclear,we need to continue to research. |
