At present,a series of nematocidal macrolide compounds were isolated from the fermentation broth of thermophilic fungi and new indole alkaloids complement with the normal temperature fungi,indicating that thermophilic fungi can produce compounds that are different from those of normal temperature fungi and have novel biological activity.Chemical investigation on two thermophilic fungi Myceliophthora thermophila THNM 055 and Thermomyces lanuginosus 200065 was performed in this thesis.Chemical investigation on M.thermophila THNM 055 strain cultured in 45 ? and PDB,led to the purification of five compounds,of which four structures(1-4)have been identified as known compounds,including a alcohols(1),a terpenoid(2),and two aromatic compounds(3,4).Compound(5)is a new compound.Compound 1 was reported for the first time in natural products.Compounds 1,3 and 4 were isolated from thermophilic fungi for the first time and tested for their attractive activity towards M.incognita.Compounds 1 and 3 show strong attractive activity at a concentration of 400?g/ml.When at a concentration of 800 ?g/ml,compound 1 displayed a repelling effect on nematodes.Chemical investigation on T.lanuginosus 200065 strain cultured in 50 ? and YGP,led to the purification of fourteen compounds,of which eleven structures have been identified as known compounds.The types of compounds are ceramide(11),long-chain fatty acid(11),naphthyl(14),pyran(17),aromatic(7),ergosterol(16-18),cyclic peptide(8,15)and a mannose(19),and all compounds were isolated from T.lanuginosus for the first time.Antibacterial activity tests showed that compounds 11 and 12 showed weak inhibitory activity against E.coli at concentrations of 20 ?g/ml and 200 ?g/ml.The metabolic maps data of crude extracts and compound samples of these two strains were established with HPLC chromatogram,which laid the foundation for the study of compounds screening.
