Re-inoculation of endophytic fungi can effectively alter the physiological and biochemical status of wine grape,or even improve fruit appearant properties of wine grapes.This provided new method for purpose shaping the qualities of the wine grape by regulating fungal community structure in grapevines.However,how to make endophytic fungi successful invasion into grapevine is the key step to achieve directional reconstruction of certain fungal endophytes contained grapevines.In this study,20 strains of endophytic fungi which isolated from the leaves of grapevines were used to infect grape calli,aseptic leaves and seedlings,field seedlings in order to investigate the technologies for successful infection and colonization of specific endophytic fungi into grapevines.More explored the impacts of specific endophytic fungi or community on the biochemistry of wine grapes.The main results are as follows:1.It is difficult to make the wine grape calli and endophytic fungi long-term symbiosis.2.Surface direct inoculation or injury inoculation can make endophytic fungi successfully invaded into grape in vitro aspetic leaves and become endophytic fungi.No significant difference was found in the effects between surface direct inoculation and injury inoculation.This method can be successfully used for studying of the infective abilities of different endophytic fungi,as well as the interactions between endophytic fungal strains,or between fungal endophytes and host grape.3.Both fungal inoculation and co-culture with fungal endophytes can make the fungal endophytes successful infection and symbiosis into tissue culture grape seedlings.4.In the field re-inoculation experiment,endophytic fungal strains,regardless the inoculation start from the cuntting or seedlings,the inoculated endophytic fungal strains were all failed to become dominant fungal endophytes in leaves of the following field seedlings.But compared to the controls,the inoculation have changed in different extents,the conmmunitiy structures of foliar endopytic fungal community structures in grapevines.5.Co-culture with endophytic fungi significantly affected the growth and PAL activities of grape seedlings.The presence of fungal endophytes during the co-culture have effects on grape seedling biochemistries such as total phenols(TPh),total flavonoids(TF),total protein(TPr)and PAL activity,but not reach the statistical significance.In the field re-inoculation experiment,the effects of different treatments on TPh,TF,PAL,TPr,TS and total titratable acid(TTA)were differentially shaped,but also not reach the statistical significance compared to the control.In conclusion,fungal endophytes community of grapevine can be artificially constructed at organ and plant levels.The re-infection of endophytic fungi or co-culture with endophytic fungi not only changed the communitiy structures of the original endophytic fungi in grape leaves,but also affect the growth and secondary metabolism of grapewine to a certain extent.This experiment has provided technologies and some foundations for the reconstruction of endophytic fungal community and the regulation of wine grapes quality characteristics.
