The blood and lymphatic vessels comprise the two largest circulatory network in higher animals,which play rolesin distributing nutrients,immune cells and antibodies,as well as hormones and other biologic active molecules into,and remove metabolic wastes from all tissues.Though connections in the jugular veins,the two vascular systems coordinate and maintain the body fluid balance.Abnormal development of the vascular system give rise to extremely annoying problems in clinical medicine.TheNetrins and its reporter UNC5B,have been reportedto playimportant roles in angiogenesis,however,roles of other members of the UNC5 receptor family in the development of circulating vascular systems have not yet been reported.In our previous work,we identified,for the first time,that UNC5C also participate inregulation of blood vascular development.At the same time,we found that UNC5C tended to affect mainly the development of veins relative to UNC5B of the arteries.The development of the lymphatic system mainly originates from the veins,so we hypothesized that UNC5C may also play a role in regulation of the lymphatic development.In this study,we investigated the role of UNC5C in the development of zebrafish lymphatic vessels using zebrafish models and studied the effects of UNC5C on lymphangiogenesis of human lymphatic endothelial cells(HLEC).We applied Crispr-Cas9 technology to generate UNC5C-/-zebrafish,RNAi and overexpression to establish modified HLEC lines.With the zebrafish in vivo and the HLECin vitrostudies,we obtain the following findings.1.UNC5C knockout in zebrafish impairs development of intersegmental lymphatic vessels and thoracic duct,showing that UNC5C plays an important role in maintaining normal development of the zebrafish lymphatic system.2.UNC5C Knockdown inHLEC inhibits their capabilities of migration and lymphatic tube formation invitro,revealing preliminarily the cellular mechanisms of UNC5C effects in lymphangiogenesis.3.Down-regulation of UNC5C,is accompanied by the down-regulation of several important genes implicated to thelymphgenesis,including PROX1,VEGFC/D,VEGFR-3(FLT4)and NRP2,suggesting that UNC5C minute directly or indirectly involved in transcriptional activationof the key transcription factor for LEC specification and of major signaling axis for lymphatic development.4.The expression of UNC5C and UNC5B may share certain complementary relationship,indicating that the UNC5 receptor family probably play an role for the developmental coordination of blood and lymphatic vascular system.Therefore in this study,we have identified important roles of UNC5C in guaranteeing the normal development of the lymphatic system,providing a new insights and data basis for further exploring the role of neural guidance molecular system in the regulation of major circulatory vascular system.
