Necroptosis and pyroptosis are two different types of programmed cell death, both have the feature of plasma membrane rupture. In this article,we studied the morphology differences between necroptosis and pyroptosis and its mechanism.Unlike necroptosis, pyroptotic cells are blebbing on the plasma membrane and produce apoptotic body-like cell protrusions(named pyroptotic bodies),before the plasma membrane rupture.Plasma membrane rupture in necroptotic cells are explosion-like,whereas in pyroptotic cells eventually become flat.Necroptosis is mediated by the protein MLKL oligomers on the plasma membrane, and pyroptosis is tightly regulated by the cleavage of GSDMD by inflammatory caspasel or caspasell. The MLKL and GSDMD sequences of N-terminal domain are very important to the plasma membrane translocation and oligomerization.
