At present,the bioactive peptides from Schizochytrium sp.has caused people's attention because of the functions with antibacterial,antipruritic,anti-inflammatory and other biological activity.However,these researches were still stayong on the laboratory stage.On the basis of the pre-period experimental research,the project focus on expanding the production of the bioactive peptides,and the production process of bioactive peptides was developed,which is prepared for the industrial production of bioactive peptides in the future.The raw material of this project was theresidue of Schizophyllum,which was extracted DHA.The residue of Schizophyllum was hydrolyzed by trypsin in the enzymolysis tank to obtain the bioactive peptides.By the procedure as following:centrifuge,ultrafiltration and desalting,and then the bioactive peptides were dried to be powder in the Spray Dryer.The bioactivity of the peptides derived from Schizochytrium sp.was tested both in vivo and in vitro.The establishment of enzymolysis production conditions:The results showed that the optimum conditions for the production of bioactive peptides as follows.Based on the single factor experiment,the orthogonal experiment was carried out to optimize the enzymatic production.The reaction conditions was that:PBS buffer system,substrate concentration of 8%,enzyme dosage of 1%,enzymolysis time of 6 h,and enzymolysis temperature of 37 ?.Under these conditions,the yield of bioactive peptides was 38.85%.The establishment of spray drying process parameters:Based on the single factor experiment,the response surface test was used to optimize the establishment of spray drying process parameters by the water content and the yield of the powder.The optimum parameters was air inlet volume of 2.51 m3/min,inlet air temperature of 182 ?,spray speed of 22000 r/min,feed concentration of 40 g/L and feed rate of 2.5 L/h.Under these conditions,the yield of the bioactive peptides was 69.17%,and the production efficiency was 553.36 g/d.The effect of the bioactive peptides derived from Schizochytrium sp.were investigated both in vivo and in vitro.The 3T3 cells were cultured for 24 h,48 h,and 72 h,respectively.It had obviously effect on the proliferation with concentration from 0.1 mg/mL to 0.001 mg/mL.However,its concentration of 1 mg/mLshowed the inhibitory effect obviously.The DSS acute intestinal inflammation model was established on the C57BL/6 mice.The in vivo experiment was carried out by intragastric administration.The bioactive peptides was measured from the four aspects of body weight change,blood in the stool,clinical score and intestinal morphology.The bioactive peptides can control intestinal inflammation,which has significant curative effects.
