In recent years,with the rapid development of domestic biological products industry,Bioreactor for animal cell culture is beginning to become popular in biological products,especially in animal vaccines,However,most of the bioreactor design in China is only blindly imitate and lack of theoretical support.A good bioreactor should have a tight structure,good and efficient mixing and mass transfer performance.The larger the size of the bioreactor,the more difficult it is to mix the mass transfer.As for the large scale bioreactor,the traditional method of bioreactor amplification has great difference with the actual situation,Therefore,in this paper,based on the ANSYS FLUENT 17.0 fluid analysis software,the structure of the 3000L level animal cell suspension culture bioreactor was simulated and optimized.In this paper,the 3000L cell suspension culture bioreactor was constructed with three layers of stirrer.First of all,in a certain speed,comparative analysis of the two types of agitator structure of the flow field of the pros and cons,select a better mixing effect of stirrer structure.Then the optimization of mixer layers,combination of different installation angles of three layer stirring paddle,on each layer of the impeller angle as a whole,one by one for each scheme of computer simulation and analysis of fluid,select the blade installation angle scheme of optimal mixing.Baffle plays a very important role in the mixing effect of the fluid,so the number of baffle and the baffle width are simulated by computer.In addition,the optimization design of the blade installation width is also carried out.After the simulation,the bioreactor structure:The ratio of height to diameter 1.7,three layer stirring paddle agitator,selection of mixing blade design by company FUL,the blade installation angle for the bottom impeller-45 degrees,-45 degrees of upper middle stirring blades,impeller-37.5 degrees,counterclockwise rotation,4 baffles were installed baffle cloth,width 80mm,length 2250mm,agitator blade width 75mm width is optimal.After optimization of the 3000L cell suspension culture bioreactor,were compared with a domestic supplier of the same scale bioreactor of the production practice of cell culture density and virus production efficiency.Under the same conditions,the cell density of the former doubled 38.1 times,the growth rate of the former was as high as 0.076 h-1 and the density of the latter increased by a factor of 29.4,and the growth rate was 0.06 h-1.The former virus constant and unit cell virus production efficiency are higher than the latter,the former HA peak higher than the latter 2.5 units.
