Soil salinization has a great influence on cotton growth and development,leading to the reduction of yield and fiber quality..It is in a great urgency to identify salt-related genes and explore their regulation mechanism for genetic engineering of salt-tolerant cotton cultivars.In previous work,we successfully identified a gene named GhACR1 in cotton by VIGS-c DNA library screening.ACR?ACT domain repeat protein?is a newly discovered type of proteins containing multi-ACT domain,which exise in plants and perform specific functions.ACRs were proposed be the specific amino acid receptor involving in amino acid signaling molecule and metabolic pathways in response to adversity and hormone stimulation,etc.However,there is little knowledge about the role ACR in response to salt stress in plant.This study preliminary explored the regulation mechanism of GhACR1 in salt responses in cotton via VIGS?Virus-induced gene silencing?.The main results were as follows:?1?In this study,GhACR1 gene including 4 ACT domain was cloned.The full length of GhACR1 is1227bp,encoding 408 amino acids.Multiple sequence alignments showed that the amino acid sequence of GhACR1 and AtACR10 are highly similar.Promoter sequence forecast found several cis elements related to drought,salt,and hot stress responses,such as ABRE,CGTCA-motif,HSE,LTR,TC-rich repeats,TCA-element,TGACG-motif,and motif?b who responses to.?2?Tissue specific expression and subcellular localization results showed that GhACR1 was t expressed more in cotton root than that inleaf and stem.GhACR1 located not only on the plasma membrane and nuclear membrane,but also in cytosol.?3?qRT-PCR Results showed that salt stress induced GhACR1 gene expression in cotton different tissues.GhACR1 gene could be also up-regulated by ABA,PEG,and H2O2,indicating that GhACR1 may participate in mutiple stress signal pathways in cotton.?4?Silencing of Gh ACR1 enhanced salt tolerance.VIGS-Gh ACR1 plants were higher than VIGS-control plants.In Arabidopsis,two acr10 mutant plants were more tolerant to salt stress,with more cotyledons greening rate and root length compared with that in WT.?5?Sixteen candidate proteins were identified by screening cotton yeast-two-hybrid cDNA library with GhACR1 as the bait.Proline transporter was selected for further study.Together,The study provides not only insight for further revealing the mechanism of GhACR1mediated salt response,but also valuable candidates for the genetic improvement of cotton salt tolerance.
