The viral enteritis of mink is an acute,highly contagious and fatal infectious disease caused by mink enteritis virus(MEV).The disease is widespread in the world.Weaned marten and young mink are most susceptible to infection and the mortality rate is as high as 60-90%.It is one of the three diseases that threaten mink farming and caused huge economic losses to the mink feeding industry.There is no specific treatment for mink viral enteritis,even interferon can not inhibit virus replication in vitro.Although there has been inactivated and attenuated vaccine aganist mink enteritis virus,inactivated vaccine immunization effect is relatively low and can not lead to cellular immunity,the attenuated vaccine has the potential risk of revenrison.The yolk antibody has its unique advantages on stable chemical properties,low cost and high yield,the far animal species biology distance relative,has become a research focus in the field of biotechnology and new drugs development in recent years.Although IgY has stronger resistance to acid and alkali and enzymatic digestion than IgG,studies show that a large number of IgY have lost their biological activity after passing through the gastric environment.It is nacessary for us to do somthing to keep the biological activity after passing the gastric environment.In this study,the F81 cell was used to proliferate mink enteritis virus,after that the layer chicken were immuned with inactivated vaccine and oil emulsion adjuvants.The yolk antibody was extractrd by the way of water dilution method.The double-antibody sandwich ELISA method was used to detectivethe special IgY which interaction with mink enteritis virus.the results confirmed the extracted IgY contains special antibodies aganist mink enteritis virus.The hemagglutination inhibition(HI)experiment and neutralization test showed that the extracted IgY could inhibit hemagglutination,and neutralize the mink enteritis virus and protect the F81 cells from cytopathy.The activity was soon lose after The IgY added into the simulated gastric fluid(SGF)and was almost lost in an instant.In the simulated intestinal fluid(SIF),the hemagglutination inhibition titers was gradually lost over time,and completely losthemagglutination inhibition titers after 4 hours.In order to maintain IgY's biological activity after gastric environment,the Eudragit 1100-55 was used as the skeleton material to make framework sustained-released tablets.The results showed that the framework sustained-released tablets could make most IgY withstand the stomach environment and quickly released in the intestinal simulation fluid,and the hemagglutination inhibition titers was detected.
