Objective:In recent years,the haze weather happened frequently in many areas of our country.PM2.5,the main component of the haze,is particularly harmful to human health.PM2.5 is a fine particulate in the atmosphere that can be inhaled by the body,and can be suspended in the air for a long time.We can understand the impact of PM2.5 high and low exposure on human health by measuring the concentrations of human serum cytokines IL-4 and IL-8of the population who under the high and low PM2.5 exposure conditions.Discuss the pathogenesis of tissues and cells injury caused by PM2.5.It can provide a theoretical basis for reducing the harm of PM2.5 to human health.Methods:1.Select the same area(Shijiazhuang city)bus drivers and office staffs as experimental research population to understand the age,gender,smoking status,basic diseases,haze exposure time.Because the working environment is different.The bus drivers were defined as PM2.5high exposure population and the office staffs were defined as PM2.5 low exposure population.The selected experimental research population are long-term residents of Shijiazhuang urban area(?2years);2.In the same period of time,venous blood were taken from the morning fasting state for 3ml.And the venous blood was centrifuged at 3000 rpm/min for 10 minutes.The upper serum was collected and stored in a the ultra-low temperature refrigerator.The temperature of the ultra-low temperature refrigerator was set to-70?;3.The serum samples of the two groups of population were melt at room temperature.The concentrations of Serum cytokines IL-4 and IL-8 in the two groups were detected by Elisa method.Results:1.There were 44 people in PM2.5 high exposure group,27 males and 17 females,with an age distribution of 44.5±4.50 years old,and20 smokers.There were 44 people in PM2.5 low exposure group,29 males and 15 females,with an age distribution of 44.3±3.64 years old,and 18 smokers.There were no significant differences on gender rate(?~2=0.196,P=0.658),age(t=0.234,P=0.815),smoking rate(?~2=0.185,P=0.667)between two groups;2.The data of China air quality on-line monitoring and analysis platform showed that the daily floating range of PM2.5 in2014 was 11~431 ug/m~3,and the daily average concentration was 116.4ug/m~3.The daily floating range of PM2.5 in 2015 was 6~391 g/m~3,and the daily average concentration was 88 ug/m~3;3.The exposure time of PM2.5high exposure group was 8~12 hours,and the average exposure time was10 hours.The exposure time of PM2.5 low exposure group was 2~4.5hours,and the average exposure time was 3 hours.The exposure time of PM2.5 high exposure group was significantly higher than PM2.5 low exposure group;4.The average levels of IL-4 in PM2.5 high and low exposure groups were 4.94±1.84pg/ml and 1.48±0.69pg/ml,respectively.The average level of IL-4 in PM2.5 high exposure group was higher than that in PM2.5 low exposure group(t=-11.689,P=0.000);5.The average levels of IL-8 in PM2.5 high and low exposure groups were26.75±10.13pg/ml and 8.74±6.14 pg/ml,respectively.The average level of IL-8 in PM2.5 high exposure group was higher than that in PM2.5 low exposure group(t=10.083,P=0.000).Conlusions:The average concentration of serum cytokines IL-4 and IL-8 in the high PM2.5exposure group was higher than that in the low PM2.5 exposure group.High exposure to PM2.5 can aggravate the inflammatory response of the human body and cause inflammatory injury in the tissues and cells of the body.Reducing the exposure of PM2.5 is beneficial to the health of the human body.
