As a model animal,mice play an important role in the development of life science.Inbred mice are widely used in genomics and genetics because of their homozygous,stable and phenotypic characteristics.In developmental biology,pharmacology,medicine,veterinary medicine and many other fields,more than 1000 inbred lines and independent distant cross groups have been successfully cultivated through long-term artificial breeding and selection.However,with the development of molecular biology and genetic marker diversity,the shortcomings of inbred strain mice such as single DNA structure and insufficient genetic diversity gradually appeared,which resulted in gene detection.Therefore,it is an important supplement to the research of wild rodents in different species or subspecies.The wild mouse subspecies are the important supplement to the existing experimental mouse resources.The development of wild animal resources and the cultivation of inbred strain mouse originated from wild rats have been paid more and more attention by scientific researchers,and gradually become the important field of research.The purpose of this study was to further inbreed LK wild mice(tentatively named LK mice)caught and domesticated in early experiment.SNP typing technique,coat color gene detection and skin transplantation were used to analyze its genetic ability.And its breeding value was discussed in order to lay a foundation for the exploitation and utilization of wild mouse resources.The results are as follows:1.LK mice were propagated and subcultured strictly according to the principle of whole sib mating.In the course of 10 years of passage,LK mice were eliminated from the first 7 groups to 2 groups,so far the C group was bred to 20 generations B group to 17 generations.The process of passage was long and arduous.It shows the phenomenon of inbreeding depression during the 10 th and 17 th generation,and the number of births and births are decreased.At present,the two groups were subcultured normally.Compared with PWK,LK had normal reproductive performance,which indicated that LK inbred strain was about to be bred successfully.2.Detection of SNP typing of LK mice: genotyping of LK mice was carried out by Hi-PCR method.The results showed that the average homozygosity of LK mice at 100 SNP sites was 98.5%,indicating that the homozygosity of LK mice basically met the requirements of inbred lines.3.Detection of coat color genes of LK mice: LK wild mice were mated with C57BL/6 mice and DBA/2 mice.The color of F1 generation of LK mice was the same as that of female parent LK mice.It was deduced that the coat color genotype of LK mice was AABBCCDD,which was basically homozygous.4.Immunological detection of LK mice: According to the national standard method of skin transplantation in inbred strain mice and rats of experimental animals,19 generations of LK wild mice in group C were transplanted with autologous and allogeneic skin on the back.The experimental results showed that the effective survival rate of skin graft was 100%,indicating that the histocompatibility gene of LK wild mice was basically homozygous.
