Porcine epidemic diarrhea?PED?with clinical signs,including diarrhea,vomiting and dehydration,is an acute intestinal infectious disease of swine that is caused by Porcine epidemic diarrhea virus?PEDV?,which has caused huge economic losses in the pig industry worldwide since the beginning of the PEDV epidemic.Vaccination is the most principal means to prevention and control of PEDV,other than biological control and prevention.Currently,PEDV vaccine remains inactivated vaccine and attenuated vaccine,which can't offer better mucosal immune so that protective immunity isn't ideal.At the same time,it's a weakness that these current vaccines cannot provide protection against new strains of PEDV and control regional epidemics.Therefore,our study focused on these weaknesses to study a safe and cheap adjuvant that enhance mucosal immune potential of these existing traditional vaccines,which is significant for preventing PEDV in our country.Research had demonstrated that cholera toxin B subunit protein?CTB?can improve immunity as an ideal candidate adjuvant of mucosal immunity.Our study assessed CTB with expression of Cholera Toxin B subunit protein?CTB?in prokaryotic expression system pET-32a,and purified and using with killed vaccine of PEDV.Finally,CTB protein with traditional vaccines assessed the immune response in mice model.This study is mainly as follows:1.Expression and purification of CTB proteinWe refer to the gene of CT from GenBank,and screen for the gene of CTB protein.After gene synthesis,a prokaryotic expression vector,pET-32a,with target gene and expression fungus was constructed successfully.The protein products expressed by IPTG inducing,and then was analyzed and authenticated by SDS-PAGE electrophoresis and Western-blot.These results show,expression of recombinant proteins,CTB protein,about30 kDa,as inclusion bodies exist in precipitates.CTB protein has been processed for degeneration,purification and renaturation.Finally,high purity protein was successfully achieved and few other proteins.2.Immune effect assessment of PEDV killed vaccine with CTB protein by hypodermic injectionPremix formulation contained PEDV killed vaccine with adjuvant AL hydrogel mixed in CTB protein.6 weeks old SPF BALB/C mice by hypodermic injection with an immunizing dose of 100?L,divided into 4 groups?AL hydrogel group,AL hydrogel and CTB group,CTB group and control group?.There are two immune for interval of a week,and orbital vein blood collection to detect IgG antibody every week during 2nd week to 7thh week after the second immunization and 3 rats were selected from every group randomly,and killed to get the intestinal tissue to detect IgA antibody during 5th week to 7th week after the second immunization.Besides,virus neutralization tests for detection of IgG and IgA antibody against PEDV and neutralization titer of antibody were determined.The immune effect of conventional inactivated vaccine with CTB protein was analyzed by subcutaneous injection of immune mice.The results show that,CTB protein can improve PEDV specific IgG in serum in mice which were significantly different compared with the other group?P<0.01?.At the same time,the neutralization antibody increases greatly compared to the other group,which is about 1:113.3?P<0.05?.However,it's invalid to enhance intestinal IgA for hypodermic injection.3.Immune effect assessment of PEDV killed vaccine with CTB protein by oral immunizationPEDV invaded intestinal epithelium,so it is needed that vaccine not only can induce pregnant sow to produce systemic immune responses but also can sufficient mucosal immune responses for giving piglet protection.Therefore,this study explored the possibility of oral immunization with CTB protein combined with PEDV inactivated vaccine.Premix formulation contained PEDV killed vaccine with adjuvant 0.4%chitosan mixed in CTB protein.Similarly,the immunizing dose of a mice is 100?L and immune methods were hypodermic injection and gavage to the 6 weeks old SPF BALB/C mice?CTS group,CTS and CTB group,CTB group and control group?.Under the same conditions stated above,virus neutralization tests for detection of IgG and IgA antibody against PEDV and neutralization titer of antibody were determined.After the oral treatment,we found that IgG level in serum is significant different at 7thh week compared with the other group?P<0.01?and its protective antibody levels were1:56.18,also higher than the other groups?P<0.05?.In addition,it's obviously enhanced in intestinal IgA level at 7th and 8th week?P<0.01?and the titer of neutralization antibody is1:33.6 which is significantly higher than the control groups?P<0.01?.In summary,in our study,we discussed that CTB protein,which not only strengthens the systemic immune combining with AL hydrogel adjuvant by injection but also promotes mucosal immune and limited systemic immune with chitosan by oral,possess excellent for immune stimulation.
