The incidence of porcine pseudorabies?PR?has increased year by year,and has become one of the most important infectious diseases that harms sow reproductive performance and seriously affects the economic benefits of pig farms,excepting the blue ear disease.Foot and mouth disease?FMD?is known as the"number one killer"in animal husbandry because of its rapid spread,wide coverage,difficult control and remedial measures.At present,the main measures to control the two kinds of viral infectious diseases that seriously threaten the healthy development of swine industry in China are vaccination,monitoring the level of PR and FMDV antibodies,making reasonable immunization procedures,and inoculating safe and efficient vaccines has become the key to the prevention and control of PR and FMD in large-scale pig farms.Therefore,the establishment of a fast and effective antibody detection technology and the development of new vaccines are of great significance for controlling and eliminating PR and FMD.In this study,the pSMK prokaryotic expression system of SUMO and histidine?His?labeled combination of small ubiquitin modified protein and histidine?His?was used to prokaryotic expression and identification of the gB full-length gene of porcine pseudorabies virus?PRV?and the structural protein of foot-and-mouth disease virus?FMDV??VP1,mutant VP2 and VP4?.The structural protein of foot-and-mouth disease was assembled and FMDV virus like particles were prepared.The results showed that the gB gene of PRV and the structural protein gene of FMDV could be soluble in the pSMK prokaryotic expression system of the combination of SUMO and His tags,and the soluble gB protein expressed in the expression had good reactivity.The mutant VP2 protein expressed in soluble FMDV and other structural proteins could efficiently assemble the virus like particle[O Type FMDV?S2093C?VLPs],which has good reactivity.The results show that the pSMK prokaryotic expression vector of SUMO gene and His tagged gene can promote the soluble expression of exogenous antigen.The structural proteins of the two viruses expressed by this prokaryotic expression system all have good reactivity and it provides technical data for further establishment of ELISA detection methods for detecting PRV and FMDV antibodies and the development of new vaccines.
