| Gene amplification molecular diagnosis technology has the characteristics of high sensitivity,good specificity,and mature technology,and is widely used in the field of in vitro diagnostics.Since the invention of PCR(Polymerase Chain Reaction)technique in 1985,researchers have continuously optimized and optimized PCR instruments with the goal of reducing reaction time required for thermal cycling,reducing instrument complexity,and simplifying detection operations.Since the introduction of the concept of micro total analysis system,a series of "chip lab" researches related to PCR have been promoted.Based on microfluidic chip technology,with different microfluidic chip types,design different functional modules that are miniaturized and integrated has become a research hotspot of next-generation PCR technology,and greatly promoted the development of microfluidic PCR technology.In order to solve the problems of traditional PCR devices such as complex structure,difficulty in carrying,slow reaction rate,and limited application scenario,an embedded microfluidic PCR fluorescence detection system based on Android system with integrated functions such as temperature control,PCR amplification and fluorescence acquisition and analysis is designed.The design starts from the overall independence and integrity of the system.The iTOP-4418 development board is the main hardware platform.The Android system is ported as a software platform.It is powered independently by a rechargeable lithium battery and a human-computer interaction platform control system is built on the development board.In terms of temperature control,a Pt heater is used to heat while a small fan is used to cool,and the temperature is adjusted by the incremental PID algorithm.The maximum heating rate can reach 30°C/s,and the maximum cooling rate can reach 10°C/s,greatly reducing the reaction time.In terms of detection,based on dichroic filters,filters and other optical devices,designed the simple and compact optical structure,and based on LED,CMOS detectors and other devices designed fluorescence detection subsystem,which enables highly sensitive real-time fluorescence detection.For scenes with higher requirements on the detection rate,such as field sites,an optional intelligent judgment function is added to automatically identify the positive reaction of the thermal cycle and stop the program,further reducing the reaction time.Without relying on external devices(power supplies,computers),the entire system can operate normally.It is easy to carry that can be applied to simple medical facilities,remote areas,and field environments where medical facilities are relatively simple,which greatly expands the application of PCR detection technology.In this paper,3 different reagents were PCR amplified and detected,and compared with the traditional PCR instrument.The experimental results show that the system can greatly reduce the total reaction time and ensure more rapid PCR amplification and detection on the basis of guaranteeing the accuracy of detection basically consistent with traditional PCR instruments,which promotes the development of PCR technology in POCT(point-of-care testing)application scenarios. |
