DNA molecules have many excellent biological characteristics.Functional DNA molecules such as nucleic acid aptamers and deoxyribozymes as new nanomaterials can be used to construct nanostructures and nanodevices with specific functions.Nowadays,DNA nanotechnology has become a research hotspot in the field of bioscience.DNA sensor and DNA logic gate are important research interests of DNA nanotechnology.The use of functional DNA molecules to build DNA sensors and logic gates has achieved multipul research results.However,with the development of DNA nanotechnology,multifunctional sensors and various logic gates have yet to be studied,more stable and effective biomolecule elements and construction methods still need to be explored.This study used the deoxyribozyme with catalytic functional properties to construct a sensor for detecting DNA molecules and DNA logic gates of low cost and easy operation.The experimental results and analysis and discussion were also given.The research content is as follows:A DNA sensor based on deoxyribozyme was designed.The three HBV DNA specific sequences of hepatitis B virus(HBV)gene were detected by this sensor.The construction of the sensor system is divided into two parts: one is the design and simulation of the sensor model.The HBV DNA sensor model was constructed by reasonable DNA sequence design and NUPACK software simulation,and the corresponding target mismatch rate analysis were carried out.The other part is the biological experiment of the sensor system.It was verified by polyacrylamide gel electrophoresis(PAGE)experiment.The sensitivity of the sensor is 100 nM.The experimental results verified the feasibility of the sensor in HBV DNA detection.NAND logic gate and NOR logic gate based on deoxyribozymes were designed.Based on the DNA sensor,the components of DNA logic gates were improved in DNA sequence and structure.Reasonable design made the input and output of the reaction system all the DNA molecules and no more than 60 bases,and obtained the four logic state models of NAND gate and NOR gate.Compared with the previous logic gates,the overall response time of the logic gate system did not exceed 3h,which minimized the construction cost of the logic gate.The design greatly simplified the operability of the experiment and showed a good experimental feasibility.The NAND gate and NOR gate are realized by PAGE experiment,which lays the foundation for realizing the connection between the DNA logic gate and constructing the continuous logic calculation system.
