Carbapenem antibiotics have been widely used in clinic because of their broad antibacterial spectrum and strong bactericidal ability,which also led to more and more resistant strains of the production.There are three main types of enzymes divided into A,B,D three categories that can be used to hydrolyze carbapenems,among which B is also known as metallo?lactamase?MBLs?and detected and reported in many countries and regions,but fewer reports were reported in wild birds.Through further research on temporal and spatial distribution of resistant bacteria carried by wild birds,we can better assess the birds in the spread of antibiotic resistant bacteria and resistance genes in the role.Samples of bird feces were collected from Nansha Wetland Park and University C ity Bay Tsui Head Wetland Park from 2015 to 2016,a total of 180 copies.10 strains of antibiotics-resistant Escherichia coli were isolated and identified in the laboratory,which carry blaNDM-5 gene and no blaOXA,blaKPC,blaVIM,blaIMwere detectedby PC R amplification and sequencing.The sensitivity of 10 strains to 17 kinds of antibiotics and the degree of drug resistance were tested by two times agar dilution method,It is shown that 10 strains were resistant to cefotaxime,ceftazidime,meropenem,ertapenem,cefoxitin,tetracycline,rifampicin and sulfamethoxazole/trimethoprim and florfenicol,moderate resistance to tobramycin,ciprofloxacin and fosfomycin moderate,sensitive to aztreonam,a mikacin,gentamicin.Followed by PFGE typing,digestion using Xba I,were digested successfully,and produced almost the same pattern.10 strains of Escherichia coli were successfully combined with thetransformation and conjugation.PCR screening and drug sensitivity test showed that blaNDM-5 was transferred.The results of S1-PFGE and Southern blot analysis showed that the blaNDM-5 gene was located in a plasmid with a size of about 50kb,it is also beneficial to the research and understanding of the mechanism of drug resistance and the way of transmission of drug-resistant genes.Wild birds have a wide range of activities and a long migration,and are likely to spread rapidly in different regions and countries by carrying drug-resistant bacteria,resulting in a wide range of resistance and threatening the survival of animals and humans.Detection of carbapenem resistant Escherichia coli in birds.The resistance phenotype and PFGE of 10 drug resistant strains was very similar,and all of them were carrying the blaNDM-5 gene.We have a preliminary understanding of the types of carbapenem genes carried by wild birds in C hina and confirm that wild birds can carry and spread drug-resistant bacteria and resistance genes through long distances.
