90% spermatophytes store lipids in seeds as their energy materials,which is an important source of the human edible oil and industry raw material.As an elementary unit of lipid storage,oil body mainly exists in the cells of the seed storage tissues,and provide energy and carbon skeleton for the seed germination and seedling growth.Meanwhile,some oil bodies can also exist in the cells of vegetative organs,as a temporary storage place of the assimilation.Besides the effect of energy storage,oil body is also close related to plant resistant and mucilage formation.PcG protein complexes,as epigenetic regulation factors,participate in many different physiological processes through change the methylation status.By use of the methods of molecular biology,biochemistry,cell biology and transcriptome,through the study of the ring(ring1a/ring1b)and sc(SWN/CLF)mutants,we found that the genes Ring1a/b and SWN/CLF belonging to PcG family were closely related to oil and oil body synthesis,and the content of oil and the number of oil body is obviously increased after knock-out ring and sc,respectively.During the process of the study,we got the following results:1.We got the mutants sc and ring with higher oil content in Arabidopsis leaves,through the screening of TLC and GC.2.The number of the oil bodies in the leaves of mutant ring and sc is increased obvious higher than the one in wild type with the observation of confocal.3.The expression levels of the genes relative to fatty acid synthesis and oil body in the mutant leaves of sc and ring were obviously higher than the ones of control.4.The transcriptome analysis results also showed that the expression levels of many genes related to fatty acid and oil body synthesis were obviously increased and majority of them could be regulated by the status of methylation.
