Siraitia grosvenorii is known as "Momordica Grosvenori Swingle".The fruit of a genus of cucurbitaceae from the dicotyledon family.Saccharide is one of the important nutrients in the Siraitia grosvenorii.However,there are no reports about the effect of Siraitia grosvenorii oligosaccharide on the proliferation of Lactobacillus acidophilus.The study is beneficial to the development and utilization of Siraitia grosvenorii and enrich products.The main results are as follows:1.Siraitia grosvenorii is analyzed in the case of dried fruit,and the extraction and purification by hot water,alcohol precipitation.Then using method of High Performance Liquid Chromatography to analysis.There are six kinds of monosaccharides in Siraitia grosvenorii: glucose,galactose,arabinose,rhamnose,xylose,glucuronic acid.2.The effect of ooligosaccharide on the proliferation of lactobacillus acidophilus was studied by plate counting method.The number of viable bacteria is increased with the increase of concentration.T he number of viable cells starts to decrease as the effect of osmotic pressure when the sugar concentration continues to increase.So the optimum concentration is 0.02g/mL.3.The study explores the metabolism of lactobacillus acidophilus in different carbon sources by plate counting method.Seven different sugars are used as the only carbon source in the culture medium for the lactobacillus acidophilus.Only polysaccharides can't be used.Lactobacillus acidophilus is good for the rest of carbohydrate metabolism.4.The study investigates the effect of polysaccharide on the proliferation effect of lactobacillus acidophilus by plate counting method.By plotting the growth curve of bifidobacteria growing in Siraitia grosvenorii oligosaccharides,glucose,galactose,arabinose,and rhamnox.The growth of lactobacillus acidophilus has the best effect which is grown in the polysaccharides,and has a longer logarithmic growth period.At the end of the experiment,the bacteria concentration is better than that of other experimental groups..At the same time,the study investigates the pH culture medium.In the logarithmic phase,the pH of lactobacillus is decreased fastest and the more acid accumulated.So the polysaccharide of the Siraitia grosvenorii increases the vitality of the bacteria which can be used as the probiotics of the bacteria.5.By plate counting method,this experiment studies the activity of lactase of lactobacillus every six hour which growing in the Siraitia grosvenorii oligosaccharides,galactose,glucose and arabinose.It was found that the activity of lactase in each group shows an increasing trend.The activity of lactase is highest when the lactobacillus grows in the lycium oligosaccharide at 18 h.6.By plate counting method,the study of the effect of Siraitia grosvenorii oligosaccharides on the increase of tolerance of lactobacillus in the gastrointestinal with glucose as a control shows that:(1)The Siraitia grosvenorii oligosaccharides can enhance the tolerance of lactobacillus which growing in the bile salts under the condition of 3mg/mL.(2)The Siraitia grosvenorii oligosaccharides can improve the gastric juice tolerance of lactobacillus significantly within 3 hours in the case of simulated gastric juice.(3)In the simulated intestinal fluid environment,the tolerance of Siraitia grosvenorii oligosaccharides to trypsin is improved within 1~4 hours.This research is to rich Siraitia grosvenorii function study,provide theoretical basis for expanding Siraitia grosvenorii tea industry application Will be made from Siraitia grosvenorii oligosaccharides processed to a product,can improve human body health,such as improving immunity,etc.
