| Super Enhancers(SEs)are large clusters of enhancers and play an important role in individual growth,immunity,metabolism and cancer pathogenesis.In recent years,the relationship between SEs and cancer has attracted much attention.Further research on SEs function in colon cancer is of vital significance to understand the occurrence and development of colon cancer and the later clinical treatment.EphA2-related tumor universal super enhancer(EphA2-SE)is selected as the experimental object in this study,which located at chr1:16498296-16509788.We found that the activity of the EphA2-SE is predominantly driven by the E1 component enhancer via the dual luciferase reporter assay in HCT-116.And we generated the knock-down and knock-out EphA2-SE cell lines by CRISPR/Cas9,then we found that target gene EphA2 m RNA expression level is significantly decreased after knocking EphA2-SE region by q RT-PCR,especially in knock-out EphA2-SE cell line.Compared with the knock-down of EphA2-SE,the knock-out can more significantly inhibit the proliferation and colony formation ability and cell migration ability of HCT-116 cells by MTT and colony formation and wound healing assay respectively.The results show that E1 component enhancer takes the main responsibility for the activity of EphA2-SE,and EphA2-SE can affect the biological properties of HCT-116 cell by regulating EphA2. |
