In this study,the kangaroo skin of Australia was used as raw material.We study the process and uses of peptide from kangaroo skin(KSP).The purpose of our research is to provide theoretical basis for practical application and marketing.In the process of KSP extraction,kangaroo skin was enzvmatically digested with six different proteases such as papain.By using ABTS scavenging ability as a detection method.The optimal enzymatic conditions for the proteases were determined by Univariate analysis.Papain is preferred.The orthogonal test of enzymatic hydrolysis of kangaroo skin by papain gives a ratio of 1:20,pH of 6.4,enzyme amount of 2%(140,000 U),and hydrolysis at 55±0.5 0 C for 3±0.15 h.After lyophilization,KSP having a good water solubility and a pale yellow color.The peptide powder yield is between 15-17%.The efficacy of the KSP was studied by free radical scavenging experiments in vitro,tyrosinase activity inhibition experiments,moisturizing ability comparison,establishing an oxidati.ve damage model of L02 cells induced by hydrogen peroxide,and antioxidant ability ex periment in drosophila.The results showed that KSP can scavenge free radical in vitro,and the KSP concentration was positively correlated with the scavenging ability.KSP has different free radical scavenging ability.Through antioxidant research in vitro,lays a good foundation of cell experiment and provides the feasibility of experiment in vivo.Inhibition of tyrosinase in vitro showed that KSP can inhibit tyrosinase activity and the inhibition type is irreversible.The in vivo cell experiments also showed that 0-0.5 ?g/?l of KSP had a certain inhibitory effect on the activity of tyrosinase in mouse B16 melanoma cells.And showed a concentration-dependent manner.The h.ydrating effects of KSP were compared with the moisturizing products oin the market.The results showed that the peptide is mild in nature and stable,can maintain moisture and supply the body's loss of amino acids and water.Among the three different peptides concentrations,the low concentration of the KSP 10 mg/ml had the best moisturizing effect.The KSP had a good moisturizing effect and can also be used as an additive for cosmetics.The purpose of study was to assess the recovery effect of KSP.First of all,an oxidative damage model of LO2 cells induced by hydrogen peroxide was established.It revealed that KSP not only significantly mitigated the H2O2-induced decrease in cell viability and improved the morphology of cells,but also reduced the MDA and the content of protein carbonyl group.Furthermore,we confirmed that kangaroo skin peptide can reduce H2O2-induced reactive oxygen species(ROS)accumulation.Taken together,our data confirmed that KSP could reduce oxidative damage on cells and could serve as a novel skin care products.In this study,different doses of KSP were used to raise drosophila.The statistical results showed that in the 20-day death of drosophila,there was a significant difference between the dose group and the control group.Indicating that the KSP can prolong the life span of drosophila.SOD and CAT showed a active-dependent relationship with the concentration of the KSP.we confirmed that KSP can reduce MDA.In order to explore the anti-aging effect,mechanism of KSP,provide theoretical basis and technical support for the further development of health products we study,the SOD,CAT and MDA.
