Obesity is a type of metabolic disease caused by factors such as genetics,lipid metabolism disorders,and improper dietary habits.The number of obese people has increased year by year as the pace of life has accelerated,and it has become an invisible killer that threatens human health.Many studies have confirmed that Akkermansia muciniphila,a bacterium in the human intestinal tract,there is a significant negative correlation between the abundance of this bacterium in the intestine and the occurrence of host obesity.In this study,the prebiotics with the best growth stimulatory effect on Akkermansia muciniphila or their combinations were screened from 27 prebiotics in a vitro experiments.An obese mouse model was constructed and the pre-screened prebiotics were administered to mice for gavage experiments.Observe body weight,fasting blood glucose,and blood lipid levels of mice.Simultaneously detect the abundance of Akkermansia muciniphila in the intestine with real?time fluorescent quantitative PCR(RT-PCR)and explore the relationship between prebiotics,Akkermansia muciniphila,and obesity.It provides a theoretical basis for the use of prebiotics to increase the abundance of Akkermansia muciniphila and thus improve obesity.In order to increase the abundance of Akkermansia muciniphila by using prebiotics,the OD 600 nm of the bacteriophage of Akkermansia muciniphila was determined on the basis of 27 prebiotics.The growth curve was drawn and the lactulose,galacto-oligosaccharide and enzymolysis egg white were selected to stimulate the growth of Akkermansia muciniphila.Based on the preliminary screening of three kinds of prebiotics,orthogonal test and two-factor combination test were conducted.The combination of 0.5%concentration of lactulose and 3%concentration of enzymatic hen egg white was best for the growth stimulation of Akkermansia muciniphila.The high-fat diet was fed to mice for 8 weeks to obtain a mice obesity model.At the same time as the 8 weeks mice were fatten,a prevention group was set up.The results showed that the Prebiotics-fed group had a certain inhibitory effect on the weight gain of the mice,and at the same time,the abundance of the Akkerman.sia muciniphila abundance in the mice was inhibited and inhibited.Increased Total cholesterol(TC)and Low density lipoprotein(LDL)levels reduced Total glyceride(TG)levels.The results showed that prebiotics had a certain inhibitory effect on the increase of body weight of mice,and at the same time inhibited the decrease of abundance of Akkermansioa muciniphila in mice.lt also inhibited the increase of TC and LDL and decreased the level ofTG.After obese mouse model was obtained,a prebiotic gavage test was performed to observe the weight loss effect of prebiotics on obese mice.The results showed that prebiotics can speed up the rate of weight loss in obese mice,increase the abundance of Akkermansia muciniphila,and reduce the levels of TC,TG,and LDL.It shows that prebiotics play an important role in the prevention and treatment of obesity and increase the abundance of Akkermansia muciniphila in the intestine.This provides a theoretical basis for improving obesity.Lactobacillus casei SY13 has a strong antioxidant activity in the intestine and may have a positive effect on the growth of the anaerobe Akkermansia muciniphila.The synbiotics consisting of SY13 and lactulose or fructo-oligosaccharide were administered to mice to observe the effect of synbiotics on abundance of Akkermansia muciniphila in the intestine of mice.The results showed that SY13 and its synbiotics could promote the abundance of Akkerma,nsia muciniphila,especially the group of SY13 and lactulose.
