This research used four different Saccharomyces kluyveri strains as experiment objects,combined with single factor and response surface methods,optimized for main components of culture medium effected by absorbance value production,optimum formulation for yeast medium was obtained.Beet molasses have spun off from the sugar process that contained rich of nutrients,can ability to use in the growing process of strains,which provides theoretical references for the future reduce cost of medium proliferation of S.kluyveri strains.Because ceramide face problem like a few sources,high cost,therefore four different numbers S.kluyveri culture expansion,extraction ceramide,determine the four different numbers S.kluyveri strains exist ceramide.The results are as follows:?1?Four different strains of S.kluyveri were cultured in three kinds of medium.Compared with the other two kinds of culture medium,the 4 strains are suitable for growth in beet molasses medium,the optimum proportion of beet molasses is 4%.Four strains in the same curing condition,there is no significant difference between the morphological structures,all to form moist,shiny,opaque rounded colony;the cells appear to be round cells of both ends.Among of the four strains,number1685 and 10955 strains'bacterial colonies and the size of cells larger than others.?2?Four strain optimum medium:No.1685:4%beet molasses,0.20%peptone,0.05%KH2PO4and 0.05%yeast extract powder;No.1892:8%beet molasses,0.15%peptone,0.05%KH2PO4;No.10847:4%beet molasses,0.20%peptone,0.15%KH2PO4;No.10955:6%beet molasses,0.15%peptone,0.05%KH2PO4.Dry matter of these four strains were 3.2g/L,11.6g/L,9.7g/L,and 10.4g/L,respectively.And it increased about 35%than before.?3?By optimizing the culture medium,four S.kluyveri strains'total lipid which extracted from them have obviously improve.Four S.kluyveri strains'polar lipids in the total lipid less than neutral lipid,and total recovery of four S.kluyveri strains were higher,more than 85%.By using thin layer chromatography?TLC?method analysis,it can be preliminarily determined four kinds of S.kluyveri contain small amount of ceramide.After the optimization of the culture medium,the size and strength of the spots can be seen in these four strains content of ceramide was obviously increased.
