Ginkgo biloba L.is the oldest relict plant of the gymnosperm left after the Quaternary glacial movement.It is used for medicinal,edible,ornamental and timber.Ginkgo biloba is a large deciduous tree with high utilization value.Ginkgo biloba fruit can delay aging,prevent cardiovascular and cerebrovascular diseases,beauty and beauty,etc.It has high edible value,ginkgo has a tall appearance,straight trunk,and has high ornamental value.At the same time,it also has a very high greening value and material value.In this experiment,the embryo and stem segments of Ginkgo biloba were used as experimental materials.Through tissue culture,the study on the factors affecting the proliferation of Ginkgo biloba,and the determination of physiological and biochemical indexes during the proliferation of Ginkgo biloba,the mechanism of the proliferation of Ginkgo biloba was revealed.Factors affecting the proliferation of Ginkgo biloba,the factors affecting the proliferation of Ginkgo biloba culture,the main findings are as follows:1.The treatment time of mercury in disinfection treatment is an important factor affecting explants.Excessive treatment time may lead to browning of explants.If the time is too short,disinfection is not easy to cause bacterial contamination.Through the disinfection treatment of two different explants of Ginkgo biloba and stem segments,it was found that Ginkgo biloba explants were best disinfected with 70% ethanol for 30 s and 0.1% mercuric chloride for 6 min.It is best to disinfect the body with 70% ethanol for 30 s and 0.1% mercury solution for 10 min.2.In the primary culture,the mature embryo of Ginkgo biloba was better and faster than the explants,and the contamination rate was significantly lower than that of the stem explants,which was more than 25% of the explants.The medium was MS medium and the induction rate reached 89.32%.The optimal storage time for the ginkgo seed embryos used for tissue culture is 2-3 months.3.The optimum medium for the primary culture of Ginkgo biloba stem by MS was 6+MS-6-BA1.5mg/L+NAA0.2mg/L+AC0.2%.The optimal selection of Ginkgo biloba proliferation was screened out.The medium was MS+NAA0.2mg/L+6-BA1.5mg/L+ AC0.2%,the proliferation coefficient was the largest,the proliferation coefficient was 3.89,and the proliferation cycle was about 40 d.4.In the process of proliferation of Ginkgo biloba buds,with the formation and growth of buds,the consumption of large amounts of nutrients is needed.The content of soluble sugar and soluble protein shows a certain regularity with the process of bud bud proliferation.Soluble sugar first rises slowly.After the rapid increase trend;soluble protein content showed a trend of decreasing first,then increasing and then decreasing.During the process of colonization and bud proliferation,the activities of POD and SOD showed the same change pattern,and the activity of POD increased first and then decreased.The activity of SOD increased first and then decreased.
