Lactic acid bacteria exopolysaccharide(LAB-EPS)is a kind of saccharides secreted during the growth and metabolism of Lactic acid bacteria.EPS has many biological activities,such as antioxidant,anti-tumor,cholesterol lowering,immune enhancement and intestinal flora regulation.In food industry,LAB-EPS could be used as natural food additives to improve the rheology,emulsification,adhesion and texture of dairy products,as well as the water retention of baked food,according to its stability and security.On the basis of the previous study in our laboratory,the extraction of lactobacillus rhamnosus BMBL0606 EPS was optimized in this study.Extraction rate,composition and antioxidant capacity were determined.Single components was separated and purified by the extracted crude EPS,and the spectral analysis and antioxidant activities were measured.And the antioxidant dropping pills were prepared by using EPS of L.rhamnosus as an effective active ingredient.The ratio of matrix and dropping temperature were optimized,and the polysaccharide content,dissolution speed and antioxidant activity were analyzed.The results were as follows.1.The production optimizations of EPS were as follows.1)59.9%of the ptotein in the fermented liquid was removed after absorbed for 4 h with 120 g/L polyamide(PA).2)The optimal parameters for ethanol precipitation of EPS from L.rhamnosus BMBL0606 were 5-fold condensation of fermenration supernatant,pH 7,4:1ethanol/fermentation supernatant ratio(V/V),and 12 h precipitation time.3)Under the optimal conditions,the EPS yield were 15.5 g/L.The EPS has 74.07%suger,1.19%protein,0.76%fat and 0.12%phenol.4)69.46%Abts.+had been removed by20 mg/mL EPS.2.EPS was purified by DEAE-52 cellulose ion exchange column,and three EPS fractions had been achieved,which were respectively named as EPS-1,EPS-2 and EPS-3.The results indicated that EPS-1 had better scavenging abilities on DPPH,hydroxyl radicals and Abts.+,while EPS-2 had weak antioxidant activity.3.Antioxidant dropping pills were successfully developed.1)The optimal ratio of matrix(PEG 2000:6000)was 1:1,dropping temperature was 75oC.Under the optimal conditions,the shaping rate of EPS drop pill was 98.1%,the average shot weight was 0.022 g,the mass discrepancy and time limit of dissolving both conformed to the regulation of China Pharmacopoeia.2)The total sugar content of dropping pills prepared under the best conditions was 17.17 mg/g.3)The half maximal inhibitory concentration(IC50)of DPPH was 227.32 mg/mL,and IC50 of OH was 43.15 mg/mL.To sum up,this study optimized the extraction process of EPS from L.rhamnosus BMBL0606,improved the quality and yield of the finished product.Then,the EPS obtained under the optimal extraction process was separated and purified,and the structure and antioxidant activity of the single components were analyzed.This is a foundation for the application of LAB-EPS in food and pharmaceutical industry in the future.The preparation technology of EPS dropping pills was optimized,its physical properties and antioxidant activity were analyzed,which provided reference for the future industrialization of large-scale production of antioxidant dropping pills.
