In this study,the species of subfamily Chironominae in Wuling Mountain region were researched by morphological method and DNA barcoding,Based on traditional morphology,the specimen was dissected,measured,described,and the previous description and record of Chironominae in Wuling mountain area from references were summarized,and the distribution catalogue of the subfamilies of Chironominae in Wuling mountain area was obtained;at the same time,the mitochondrial c oxidase subunit 1(COI)gene of 30 species of Chironomidae in Wulingshan area was amplified by PCR,and a total of 113 sequences were obtained.MEGA 6.0 software was applied for sequence analysis and comparison,the genetic distance(genetic distance)were analyzed based on kumara-2 parameter model,base composition(base composition)and transition/transversion ratio(ts/tv ratios)were statisticed,the phylogenetic tree was constructed by the neighbor-joining method(NJ)and the maximum likelihood method(ML).The results of the study further clarified the systematic classification status of Chironominae,filled the blank in the systematic research of subfamily Chironominae in the region.The acquisition of molecular data has expanded the barcoding database of subfamily Chironominae and provided a scientific basis for the phylogenetic analysis of Chironomidae.The main conclusions as follow:(1)16 genera and 50 species of subfamily Chironominae had been recorded in wuling mountain Area.Two new genus records were added,including: Genus Dicrotendipes Kieffer;Tanytarsus v.d.Wulp;12 new recoreded species were added,including: Dicrotendipes flexus(Johannsen),Dicrotendipes pelochloris(Kieffer),Chironomus(Chironomus)cingulatus Meigen,Chironomus(Chironomus)circumdatus Kieffer,Chironomus(Chironomus)javanus Kieffer,Chironomus(Chironomus)kiiensis Tokunaga,Demicryptochironomus(Irmakia)constrictus Yan,Tang & Wang,Glyptotendipes tokunagai Sasa,Kiefferulus barbatitarsis(Kieffer),Polypedilum(Polypedilum)nubifer(Skuse),Tanytarsus formosanus Kieffer,Tanytarsus takahashii Kawai & Sasa.Through this study,it was found that there were abundant species diversity of Chironomidae in this area,from another aspect,it was also proved that Wuling Mountain area is very rich in biodiversity.(2)In this study,113 sequences of coi gene were obtained,including 102 of Chironominae,6 of Orthocladiinae and 5 of Tanypodinae.Based on the distance matrix,the intraspecific and interspecific genetic distances of some subfamilies of Chironominae were analyzed in the experiment,the average interspecific genetic distance of all species is 16.89%;the maximum interspecific genetic distance is 27.9%,the minimum interspecific genetic distance is 12.1%,the maximum genetic distance within the species was 5.1%.(3)The phylogenetic tree was constructed by using the corrected distance kumara-2 parameter model of MEGA6.0,and the molecular species were successfully matched with morphological species.
