The Brachyury(T-box)gene plays an important role in regulating mammalian embryonic development,especially the formation and development of the notochord.Mammalian embryos lacking Brachyury protein cannot develop normally,and the tail phenotype of the animal is unnormalized.In order to reveal the potential interaction between the Brachyury(T-box)gene and the short-tailed traits of Hulunbeier short-tailed sheep.This experiment used the Sanger sequencing technology to compare the genotypes of the Brachyury(T-box)gene of the Hulunbeier short-tailed sheep and their parental and Ujimqin sheep.Using X-ray photographs and making the tail vertebra specimens to compares three characteristics of sheep's tail vertebraeand and the tail vertebrae specimens.Detection of methylation status of Brachyury(T-box)gene by bisulfite sequencing PCR method.The results are as follows:1.The self-crossing progeny of the Hulunbeier short-tailed sheep had the same genotypes which their parental since the initial codon the c.G333C,c.G334T locus,whereas different from the Ujimqin sheep.2.The length of the Hulunbeier short-tailed sheep caudal vertebrae was shorter than that of the Ujimqin sheep,and there are deformities.3.The degree of promoter sequence methylation of Brachyury(T-box)gene in Hulunbeir short-tailed sheep and Ujimqin sheep was 2.50%and 8.33%respectively.There was significant difference at the-97-,-118-site of the CpG island prediction site.The degree of the second exon sequence methylation of Brachyury(T-box)gene in Hulunbeir short-tailed sheep and Ujimqin sheep was 43.48%and 27.39%respectively.There was significant differenceat the-167-,-175-,-188-,-224-,-245-,-299-,-314-site of the CpG island prediction site.This study combined with the previous research results of the laboratory to further explore the molecular mechanism of the short-tailed phenotype of Hulunbeier short-tailed sheep.It was found that the Brachyury(T-box)differential gene can be stably inherited,and the epigenetic information was analyzed to prepare for the verification of differential genes.This will provide a theoretical basis for the functional study of the Brachyury(T-box)gene and the molecular breeding of the Hulunbeier short-tailed sheep.
