Rice ALOG(Arabidopsis LSH1 and Oryza G1)gene family has 10 members(G1,G1Z1?G1L9),among which three genes,G1,G1L5 and G1L6,have been reported to have functions,and they are involved in the development of rice spikelet flower in different ways.This experiment focused on the functions of ALOG family members except for G1,G1L5 and G1L6,and on this basis,the mechanism of G1L1 in the development of stigma in rice was studied.In this study,CRISPR/Cas9 gene editing technique was used to knock out all members of ALOG gene,obtain their knockout plants,and construct overexpression lines of other 7 members except G1,G1L5 and G1L6.No phenotypes were observed in the knockout and overexpression lines except the reported phenotypes.Since the sequence of ALOG members is similar,it is speculated that there is redundancy in their functions,so the 7 genes may not cause developmental defects by themselves.We investigated the redundancy of these genes through chimeric suppressor gene silencing technique(cres-t).Specifically,one transcription factor was fused to the motif inhibitory domain(SRDX)(with 12 amino acids added at the end),allowing the fusion protein to competitively bind to the regulatory region of the transcription factor and its redundant factors,thereby inhibiting the transcription factor and its redundant factors.We constructed chimeric silencing gene strains of G1L1 and G1L3,G1L1-SRDX and G1L3-SRDX,and observed that G1L1-SRDX transgenic plants showed defects in stigma development,degenerated abnormal stigma development and reduced fertility.This indicates that G1L1 and its redundant factors are involved in stigma development.Similarly,we also found that G1L3 is also involved in the growth of rice plant height.Further studies on ALOG family members showed that G1L1 was a transcription factor and expressed in the nucleus.G1L7 has no transcriptional activation activity and is expressed in the nucleus,cytoplasm and cell membrane.In this study,we used molecular biology techniques to reveal that G1L1,a member of ALOG family,was involved in the redundant development of rice style.It provides clues for further study on the mechanism of rice style development.
