Henan is located in the middle latitude zone,It's the central plains of The Central Plains,the terrain is high in the west and low in the east,the surface morphology is complex and diverse;The diversity of soil fungi varies with soil types.Soil Hyphomycetes are an important part of soil fungi,which are abundant in species and widely distributed.Henan province is a major grain province.It belongs to the intermediate transitional zone of climate,fungal species are abundant in resources,but the research and excavation of fungal resources are not sufficient.In order to develop and protect the species resources of fungi in China,excavate new species,further improve the data of Hyphomycetes in China,and develop soil Hyphomycetes with biocontrol activities,it is of great significance to the diversity research of hyphomycetes(1)Collection of soil HyphomycetesIn order to discover rare species of soil Hyphomycetes fungi and screen strains with biocontrol activity,soil samples were taken from 81 counties and cities in Henan province and parts of Shandong,Hubei,Inner Mongolia,Jiangsu,Yunnan and Anhui provinces.The samples involved farmland soil,wetland soil,grassland soil,park soil,valley soil,forest soil,orchard soil,bonsai soil and other ecological types.Total 827 soil samples,collected strains 743 strains,belonging to 26 genera,Bayesian method and Maximum Likelihood method to construct phylogenetic tree,by multigene phylogeny data and morphological identification more 35 known species and 15 new species,including 5 new record genus and 8 new record species(2)Preliminary screening of biocontrol activitySoil samples of different ecological types were collected from various regions of Henan province,and soil moniliaceous Hyphomycetes fungus were isolated.Strains of Myrothecium-like,Stachybotrys-like,Clonostachys and Acremonium were selected.A total of 66 strains were selected,and 5(HN17434,HN17556,HN17216,HN17506 and HN17479)strains were selected from the laboratory plate screening,plate rescreening and metabolic liquid culture of specific medium.The soil biocontrol fungi that had inhibitory effect on vermiform fungi of the Root Rot in wheat,inhibition ratio were all over 60%;In this experiment,genomic DNA was extracted from the strain with a good final effect,and then PCR amplification was conducted on ITS1 and ITS4 universal primers,and morphological characteristics were observed and identified,HN17434:Acremonium pteridii,HN17556:Albifimbria sp.,HN17216 Paramyrothecium sp.,HN17506:Paramyrothecium hubeisis,HN17479:Stachybotrys henansis.
