Sphingosine metabolites play an important role in regulating cell activity and maintaining the balance of sphingolipid metabolism.They play an important role in cell growth and development.Sphingosine kinase(SK)is an important regulator of sphingolipid balance and plays an important role in cell proliferation,migration,invasion and angiogenesis.SKI has the function of promoting cell survival and proliferation.SKI may be involved in the process of embryo implantation,which mainly involves cell proliferation,migration and angiogenesis.In this study,we established normal pregnancy model,false pregnancy model,artificial decidualization model,delayed implantation model and activated implantation model in mice.The expression of SKI in uterus tissue of each model was detected by immunofluorescence method,and the expression of SKI in normal pregnancy model of mice was detected by Western blot.The decidualization of mouse uterine stromal cells was induced in vita.The expression of SKI in decidualized cells was detected by real-time quantitative PCR,and the correlation between SKI expression and decidualization was further determined.Immunofluorescence results showed that SKI was mainly expressed in stroma area at 1-4 days of gestation,but not in uterine cavity and glandular epithelium.SKI was mainly expressed in secondary decidua area at 5-8 days of gestation.Subcellular localization showed that SKI was expressed in the cytoplasm of decidual cells.SKI was mainly expressed in the matrix region in the model of pseudopregnancy in mice.The expression of SKI on the 5th day of pseudopregnancy was lower than that on the 5th day of normal pregnancy.SKI expression was also detected in delayed implantation model and activated implantation model,which were similar to that on day 4 and 5 of normal pregnancy,respectively.SKI expression was detected in both artificial decidualization model and mouse decidualization cells induced in vita.Western blot results showed that the expression of SKI increased gradually from 5 to 8 gestation.Real-time quantitative PCR results showed that SKI expression in decidualization cells cultured for 72 h and 96 h was higher than that in control group.In conclusion,the expression of SKI in mouse uterus showed time-space-specific changes.The expression of SKI in early pregnancy was related to activation of blastocyst and decidualization in vivo and in vita.It indicated that SKI participated in embryo implantation in early pregnancy of mice.
