In mammals,inner ear is one of the most complex structures,and its structure has an obvious differentiation during the development.The protein family,LMO is made of 4 transcriptional regulatory factors which take different effects in cell differentiation.Lmo1 was expressed in the cochlea during the embryonic period of mice,which may have the interaction with transcription factors such as GATA3,ISL1,ISL2,which we are studying now.It is benefit for understanding the regulatory network mechanism of early cochlea development to confirm the influence on cochlea development from Lmo1.We researched on the effect of cochlea of cochlea development and hearing of adult by Lmo1 knock out in mice.We detected the expression pattern of Lmo1 by in-situ hybridization,and knock in report gene LacZ to knock out Lmo1,observed the influence of the organ of Corti after knock out Lmo1 by immunofluorescence,we mainly observed hair cells,support cells and spiral ganglion,in order to confirm the effect of process of cochlea development by knocking out Lmo1 in mice.Lmo1 began to express in cochlea at 13.5 days of embryonic.The results suggested that there were no evident effects on hair cells,support cells and spiral ganglion cells.Besides,the ABR results showed that mutant were not obvious hearing loss compared with the wide type.And,we found that not all of Lmo4 knock out can generate abnormal organ of Corti,about 30% of the Lmo4 knock out could lead to OC exogenous,but there are shorter cochlea duct and cochlea structure deformation when Lmo4 was knocked out.No obvious change was viewed on the organ of Corti after gene Lmo1 and Lmo4 double knocked out.
