As we enter the post genome sequencing era,the need for technologies that allow high-throughput,high-sensitivity and high-specificity immuno-detection becomes apparent in bio-detection and diagnose.Beads on beads(BOBs)structured particles have become more and more popular in the application of diagnose as potential bio-carriers for special surface structure and high surface area.For design high performance application based on BOBs structure particles,it is urgently necessary to understand the influence of surface structure of the carrier on biomolecules such as protein in application.Firstly,three kind particles with precision surface structure were designed.One was carboxylic magnetic particles(MPC)based on magnetic particles(MPs)with smooth surface,and the other two were based on BOBs particles with the same surface structure but different surface functional group distribution.Then,the capability in recognition immune complex of antibody immobilized on these three kind particles were studied via ELISA detection for unveiling the regulation and mechanism of the influence of surface structure on protein bio-activity.For this purpose,raspberry like carboxylic silica particles on amino magnetic particles(SCMPC)with precision surface structure and amino group modified on host particles was prepared basde on carboxyl group modified silica nanoparticles and amino group modified MPs via EDC/NHS process.Then,carboxylic silica particles on carboxylic magnetic particles(SCMPC)was prepared by the surface modification of SCMPN and had the same surface structure to SCMPN.Basic physicochemical properties of this three kind particles was studied.And then,anti-?-h CG antibody was immobilized onto three kind particles to study the capability in recognition immune complex in PBS buffer and 30%(v/v)serum system respectively as model protein.The result showed that antibody immobilized on SCMPN presented the highest bio-activity than MPC and SCMPC,and SCMPC was the lowest among these three kind particles in ELISA and flow cytometry immuno-detection assay.In addition,the surface curvature of carriers and assembling structure in BOBs particles were also found to have great influence on antibody bio-activity.Antibody immobilized on carrier with big surface curvature remain much more bio-activity than small surface curvature.And the assembling structure in BOBs particles was also found to have negative influence on antibody bio-activity immobilized on host particles.The result provided some theoretical guidance for design and preparation bio-carrier with high detection performance to some extent.
