PCR(polymerase chain reaction)is one kind of important means researching DNA and genes.In recent years,PCR microfluidics based on MEMS technology have obtained a rapid development due to its advantages of little sample consumption,high efficiency and low-cost.In the future,this kind of chips must be one of common means in fileds of clincial medicine and life science.The purpose of this research is to build a system that can amplify different kinds of sample in different channel with different temprature conditions at the same time.The main contents and results of this thesis are as following:1.To provide more uniformity of temperature distribution,a new heater with optimized electrode pattern based on previous serpentine heater is presented.FEM simulation and infrared thermometer test verified that the new heater can provide uniform temperature distribution in a larger area where temperature fluctuation is within 1℃.A separable PCR chip,with reusable electrodes part and disposable PDMS-glass bonded chamber part,is fabricated using MEMS technology.2.The establishments of multi-channel temperature control system based on ARM include:(1)Building up hardware palatform.(2)Transplantation and application of UCOS operating system.(3)Implementation of temperature-adjust algorithm.(4)The implement of PC-based softerware.3.PCR amplification experiment was carried out on the fabricated separable PCR chips and multi-channel temperature control system described in this thesis.Compared with results of conventional PCR,PCR was successfully amplified on the PCR chips with a 239 bp target of DNA.
