Morchella lives in soil,which is closely related to Morchella.In this study,the chemical properties of cultivated and uncultured Morchella esculenta soils in eight areas of Sichuan Province and the mineral elements in the ascocarp of Morchella esculenta are analyzed.On this basis,the diversity of microbial communities in cultivated soil of Morchella esculenta in eight areas is studied by high-throughput sequencing.The results provide valuable references for revealing the soil characteristics of Morchella esculenta cultivation for nutrition and structure characteristics of main microbial groups during the development of ascocarps.The experimental results are as follows: 1)Soil chemical properties and ascocarp element analysis of Morchella esculentaThere are great differences in soil chemical properties in 8 regions.The soil pH of cultivated Morchella is between 5.14 and 7.40,and that of uncultured Morchella is between 5.23 and 6.88.The soil pH of cultivated Morchella is higher than that of uncultured Morchella.The contents of OM,OP,TCa,TFe,TK,TMg,TMo and TZn in cultivated Morchella soil are generally higher than those in uncultured Morchella soil,and the contents of AN in cultivated Morchella soil are lower than those in uncultured Morchella soil.The average content of K is the highest in ascocarp of Morchella esculenta,followed by Fe,Mg,Ca,Zn,and the average content of Mo is the lowest.The variation coefficients of six elements ranged from 0.11 to 0.87,and 0.1?CV?1,which belongs to moderate variation.The coefficient of variation of Fe is the hishest,which is 0.87,and the coefficient of variation of Zn is the lowest,which is 0.11.OP is positively correlated with ascocarp K,Ca is positively correlated with ascocarp K.2)Analysis of soil bacterial diversity of cultivated Morchella esculentaAnalysis of soil bacterial diversity of cultivated Morchella esculenta,at the phylum level,the most abundant phylum of soil samples in 8 regions is Proteobacteria,while the first four phylums are Proteobacteria,Bacteroidetes,Acidobacteria and Actinobacteria.At the genus level,the most abundant genera of soil samples are different in eight regions.Pseudomonas are found in Mianyangyouxianqu,Sphingomonas are found in Chengduxinduqu,Qingchuanbanqiaozhen,Ya'anhongxingcun and Ziyang,Alistipes is found in Qingchuanqimaxiang,Massilia is found in Luzhoushuiweizhen,and Thiobacillus is found in Jintanghuangjiagou.At the species level,soil samples from eight regions contain B.niacini,P.vancouverensis,B.elkanii and R.ferrireducens.3)Diversity analysis of soil fungi of cultivated Morchella esculentaDiversity analysis of soil fungi of cultivated Morchella esculenta,at the phylum level,ascomycetes are the most abundant soil samples in Ziyang,Chengduxinduqu,Mianyangyouxianqu,Qingchuanqimaxiang,Qingchuanbanqiaozhen,Luzhoushuiweizhen and Jintanghuangjiagou,while mortierellomycota are the most abundant soil samples in Yaanhongxingcun.At the genus level,the relative abundance of Morchella in Qingchuanqimaxiang and Luzhoushuiweizhen are 71.29% and 75.56%,respectively;the relative abundance of Morchella in Mianyangyouxianqu,Chengduxinduqu and Jintang huangjiagou are 34.03%,40.43% and 29.54%,respectively;the relative abundance of Morchella in Ziyang,Qingchuanbanqiaozhen and Yaanhongxingcun are extremely low.At the species level,the common species in the soil samples from eight regions is M.sextelata,M.importuna,T.pleuroticola,O.brassicae,F.oxysporum,U.virens,T.phialosperma.
