Chlorella is considered to be the best raw material for the production of biodiesel because of its high fat content,easy mass cultivation,small footprint and high fat yield per unit.In this paper,the laboratory is a new separation of green algae DX01 growth situation and three periods of several kinds of main nutrients:intracellular total sugar,protein,and total fat were determined and analyzed,studied the change of pH,sodium carbonate and sodium nitrate on algae DX01 biomass and intracellular biochemical components,explore the industrial requirements,and different levels as the feasibility of biodiesel raw material.The results show that the green algae DX01 cells in different growth period of total sugar,protein,and total fat content is respectively:19%to 34%,11%to 27%and 11%to34%.First,different pH have different effects on DX01 several biochemical composition,pH value of 7.5 for intracellular accumulation of total sugar,content reached 34%of the dry weight of the cells,the accumulation of total fat and total protein was not high;pH of 8.0 for intracellular accumulation of total fat,reached 33%of the dry weight of the cells in plateau,is not conducive to the accumulation of total sugar and total protein;pH of 8.0 conducive to the accumulation of intracellular proteins,reached 27.5%of the dry weight of the cell.Although the pH value of 8.5 total sugar,protein and fat content is not high,but the biomass is big,therefore,from the many intracellular biochemical composition point of view,pH 8.5have more advantages than the other group.Secondly,sodium carbonate and sodium nitrate on algae DX01 growth and biochemical composition and the effect of the experimental set up different culture conditions,namely the sodium carbonate concentration gradient:0 g/L,0.5 g/L,1.0 g/L,1.5 g/L,2.0 g/L;Sodium nitrate concentration:0 g/L,0.125 g/L,0.25 g/L,0.375 g/L,0.5 g/L,pH to 7.5.Were measured at different times and different concentration of sodium carbonate and sodium nitrate algae DX01 dynamic change rules of intracellular biochemical components.Finally,optimized by response surface method of green algae DX01 total lipid extraction technology conditions for comprehensive analysis,finally get the best technological parameters for:chloroform/methanol was 0.6,extraction temperature 54 ~oC,the extraction time was 87 min.Under the process parameters,the actual measured algae DX01 extraction yield of 26.22%of total fat,and the model prediction is about 1%26.35%.
