Tumor necrosis factor?converting enzyme?TACE?belongs to the ADAM protein family of disintegrins and metalloproteases.TACE plays a role in the processing of tumor necrosis factor alpha?TNF-??and the release of a diverse variety of membrane-anchored cytokines,cell adhesion molecules,receptors,ligands,and enzymes.It have confirmed that iRhom-TACE pathway regulates Drosophila sleep.Either irhom or TACE mutant flies exhibit narcolepsy-like phenotype.In order to investigate the downstream protein of the iRhom-TACE pathway,itraq techniques and bioinformatics methods were used to detect proteome in taceko and iRhomkoo Drosophila brain?head?.We analyzed the interactions between proteins using.Connectin?Con?is one of the important molecules affected by mutation in tace or iRhom.Previous studies have already showed that Con is a molecule that mediates homophilic cell adhesion,but its function is unknown.To analyze the in vivo biological function of Con,we constructed Con-Gal4 transgenic lines using CRISPR/Cas9 technology.We used the Con-Gal4 transgenic lines studied the temporal and spatial expression patterns of Drosophila Connectin.Our results exhibit that Connectin is not only expressed in the Drosophila larval motor nervous axons and body wall muscles,but also in the larval sensory neurons and adult mushroom bodies.Moreover,with the development of Drosophila,the expression of Con is different both in the brain and muscle,so Con may play an important role in the development of Drosophila.
