Establishment Of In Vitro System Of Fox Cells And Analysis Of Their Biological Characteristics

Corsac fox,also known as Dongsha fox,plays an important role in ecology such as maintaining the food chain.Presently,the population of corsac fox is sharply reduced.This experiment was carried out to help to protect and develop this species resource effectively,and keep ecosystem balance.Studies on cell culture,establishment and biological characteristics of corsac fox have not yet been reported.In this experiment,the trachea,hose and ear skin tissues of corsac fox were selected and then using tissue block adherence method to construct primary cell lines,and two resources of tissue cell systems,trachea fibroblast and cartilage fibroblast,were successfully established.The two cell lines both conformed to the basic characteristics of fibroblasts,which exhibited a cell growth of adhered and typical spindle and polygon.The biological characteristics of these tissue cells from tworesources were analyzed.The adherence rates of the two tissue cells and the survival rates before and after cryopreservation were measured,.The growth curve was drawn and the result showed a S-shaped growth curve.The transfection rate of liposome transfected green fluorescent protein plasmid(PEGFP-C1)was more than 20%.Chromosome samples were prepared by conventional low osmotic drip method.Karyotype and G-banding analysis showed that the diploid(2n=36)of metaphase chromosome accounted for 84% of the total cells,and there were 17 pairs of autosomal showed a morphological type of 1st+10m+6sm,the other pair was the sex chromosome X with a morphological type M.In this study,two fibroblast cell lines and their culture system were successfully established.The chromosome number of these cells was detected and determined to be in line with the species standard,and for the first time to draw up the G-band map of the chromosome,which laid a foundation for the establishment of the genetic resource pool and the protection of the genetic resources.
