The most effective way that control the spread of pathogens is preventive inoculation with vaccine.The spread of porcine circovirus type 2(PCV2)has caused huge damage to the industry of pig worldwide.It is the main mean of preventing and controlling of pathogen infections that use of vaccines for prophylactic vaccination.A series of diseases caused by Porcine circovirus type 2(PCV2)have made irreparable damage to pig industry worldwide.Nowadays,many commercial porcine circovirus vaccines have defects of inefficiency and insufficient immunogenicity,and can't play the role of immune protection.And that the existing adjuvant of vaccine has non-negligible side effects.Therefore,it is no time to delay that develop and search new vaccine adjuvants that is safe and effective.Inulin(INU)is mainly derived from plants and obtained easily,and it is low in cost.It is the most important that inulin has no toxic and no side effects,and it is widely used in the food industry.It has the effect of life cultivation and health preservation,and has been proved by the laboratory to have vaccine adjuvant activity.In this paper,a novel porcine circular subunit vaccine was prepared by covalently linking inulin(INU)and porcine circovirus type 2(PCV2)subunit vaccine by chemical coupling technology.The adjuvant-antigen conjugate subunit vaccine PCV2-INU was prepared by chemical modification method,and the conjugate was characterized by SDS-PAGE,Size Exclusion Chromatography,Endogenous Fluorescence Spectroscopy,and Thermal Stability Measurement.The results of analysis suggest that the relative molecular mass of conjugate product is significantly increased,and it has good binding effect.The study of immunology have shown that the use of inulin as an adjuvant to modify the PCV2 subunit vaccine,the immunogenicity of the vaccine can significantly improve and induce a strong Th2 type immune response,which provided effective immune protection for organism.In addition,Inulin can't induce organism to produce anti-inulin adjuvant the specific antibodies,and no any side effects about organism,it can be used with confidence.Therefore,it is a feasible,safe and effective strategy to modify the PCV2 subunit vaccine with inulin that can improve its immunogenicity.
