B-cell epitope identification plays an important role in vaccine development and disease diagnosis,but locating the epitopes using traditional methods is time consuming and expensive.Computational prediction of epitopes using affinity-selected peptides is an economical and effective approach.In this paper,we focus on the prediction of epitopes using affinity-selected peptides,doing much research work in the target-unrelated-peptide(TUP),new epitope prediction algorithm and tool and the B-cell standard test datasets.By studying the experimental process and method of traditional phage display,and comparing and analyzing the existing TUP screening tools,a plastic surfacing binding peptide(PSBP)recognition online service called PSBinder was developed.The service adopts the support vector machine algorithm,and selects the optimized amino acid component information as the feature to construct and has good performance.You can get the service at http://i.uestc.edu.cn/sarotup/cgi-bin/PSBinder.pl?Based on the research of the evaluation data of the B-cell epitope prediction algorithm based on mimotope,further excavation processing was carried out on the basic original research work.We have compiled a set of standard test data sets containing template complexes,standard epitopes,and mimotopes.By studying the implementation methods and core algorithms of existing B cell epitope prediction tools,and aiming at the deficiencies of existing tools,a new B-cell epitope prediction tool was developed using a new idea and algorithm,and we implemented it as the plugin of PyMOL software.With PyMOL's powerful molecular 3D structure display capability and flexible operation capability,the tool has become a tool with cross-platform,fast running speed,data guarantee,high prediction accuracy,flexible display and operation.In summary,this research has developed a new TUP recognition tool and epitope prediction tool through the research of the whole B cell epitope prediction process,and updated the B cell epitope standard test dataset.This study has a driving role in phage display noise recognition,vaccine preparation and disease diagnosis,and has important scientific significance and application prospects.
