Microfluidic sensor is a very important biochemical analysis tool.The use of microfluidic sensor to detect trace substances in liquids is of great significance in biomedical,environmental monitoring,and disease detection.Protein and DNA play an important role in the human body.Some diseases?such as hepatitis B,tumor,nephritis,etc.?can cause abnormal protein or DNA content,so the trace detection of these two substances is of great significance.In this thesis,the detection methods of protein and DNA were investigated,and the advantages and disadvantages of various detection methods were compared.This thesis builds a test system based on the detection principle and designs an experimental plan.Then the instrument noise,the state of the left and right pipelines,the concentration pollution and the instrument cleaning ability were tested,which proved that the instrument is stable and reliable.The flow rate pattern was tested and compared,the appropriate flow rate mode was selected and the appropriate test period was determined.The content of bovine serum albumin and the content of calf thymus DNA were tested.In the test of bovine serum albumin content,the effect of the rhodamine B concentration on the test and the influence of the excitation light intensity on the test were studied.The interference factors were also tested and analyzed.The interference factors mainly included BSA samples and NaCl interference and thermal denaturation interference.Through experiments,it was finally determined that under suitable conditions,the detection limit can reach 1×10-12mol/L,and the linear detection interval is 1×10-9mol/L1×10-12mol/L.In the test of the ctDNA content,and the effect of fluorescent probe rhodamine B concentration on the test was studied.The detection limit(1×10-12g/mL)and linear detection interval(1×10-9g/mL1×10-1212 g/mL))were determined..Thermal denaturation tests were also performed to demonstrate that thermal denaturation has an effect on the binding of Rhodamine B to DNA.Tests show that the performance of the metallic-capillary is superior,and it has a good application prospect in the field of trace detection.
