Study On Morphological Structure And Bacterial Community Of Intestine In Melophagus Ovinus

In this study,were used the super depth of 3d microscopy system and SEM to observe external morphological characters of the Melophagus ovinus which is the ectozoa of sheep(from Gansu),were analyzed 18 S genes and mitochondrial cytochrome oxidase subunit I gene(COX-1).Then,the bacterial composition of female and male M.ovinus in the midgut and pupa were analyzed by PCR-DGGE and NGS.Morphological observation showed that the body of M.ovinus was divided into three parts: head,chest and abdomen.M.ovinus was an entirely wingless brown fly,no halteres.The head was short and broad flat,M.ovinus had piercing-sucking mouthparts,reduced compound eyes and thumbnail antennae,no ocellus.The chest was not segmented,had three pairs of legs and four pairs of stegmas.Sequencing results shows that the length of the 18 S gene sequence was 522 bp,which had a high hormology of 99% compared to Geckobia(NO.KY224149.1);The length of the COX-1 gene sequence was 770 bp,which had a high hormology of 99% compared to GenBank(NO.EF531209.1).The result of PCR-DGGE reveals that the dominant bacteria of female and male M.ovinus in the midgut and pupa were Bartonella,Arsenophonus and Wolbachia;Microflora of female and male of M.ovinus in the midgut were similar to the pupa.The result of NGS shows that female and male M.ovinus in the midgut and pupa had 10OTUs?11OTUs?12OTUs respectively.Among them,8 OTUs were shared between female and male M.ovinus in the midgut and pupa.Except these OTUs,they were 0 OTUs?2 OTUs and 12 OTUs specific to female and male M.ovinus in the midgut and pupa respectively.The highest abundance bacteria are different in female and male M.ovinus and pupae,Arsenophonus is the greatest abundance on female adult M.ovinus in the midgut,it is close to 55%,Bartonella is the greatest abundance on male adult M.ovinus in the midgut,it is close to 60%,Wolbachia is the greatest abundance on pupa,it is close to 50%.
