In recent years,Aeromonas veronii(A.veronii)has become a new type of pathogen for human,,and has become more and more important in public health safety.It has been listed as a key subject for water quality and food safety by many countries.Most of the researches on A.veronii at home and abroad focus on the identification and identification of common virulence factors,but there are few studies on its pathogenic mechanism.It has been reported that Cpx two-component systems are widely present in Gram-negative bacteria and play an important role in regulating bacterial pathogenicity,but their research in A.veronii has not been reported.The function of related genes in the Cpx two-component system,speculated to play a regulatory role in A.veronii,helps us to further understand the pathogenesis of A.veronii.The cpxA and cpxR genes in the two components of A.VTH0426 Cpx were constructed by gene knockout technique.The biological characteristics of wild plants were compared and found to be ?cpxA and ?cpxR and wild strain.Compared with the bacterial growth state,hemolytic,physical and chemical properties and drug sensitivity test,In the stress tolerance experiment,the simulated high osmotic pressure and oxidative stress showed that the tolerance of ?cpxA was lower than ?cpxR and both were significantly lower than that of wild strain,and the tolerance of ?cpxA under simulated acidic conditions.Above ?cpxR but still lower than wild strain,the tolerance of ?cpxR was higher than that of ?cpxA and both were higher than that of wild strain under simulated alkaline conditions;?cpxA and ?cpxR were found in the detection of biofilm formation ability.The growth time of the biofilm formation was gradually reduced compared with the wild strain;the exercise ability test showed that the swimming diameter of the wild strain was 3.43 times and 3.68 times of ?cpxA and ?cpxR;In the pathogenicity test,?cpxA and ?cpxR on zebrafish the LD50 increased by 14.5 times and 30.8 times,and the LD50 of mice increased by 6.1 times and 11.6 times,and the adhesion and invasion ability of EPC cells decreased by 1.52 times and 2.28 times.The virulence to EPC cells decreased by 1.97 and 2.89 times.Initial exploration of the function of cpxA and cpxR genes.Combining the results of the study of cpxA and cpxR gene functions in two components.Analysis shows that: The Cpx two-component system is involved in the regulation of A.veronii TH0426 stress tolerance,biofilm formation,and pathogenicity.In summary,we envision whether the Cpx two-component system can be regarded as an important virulence factor,further analyzing the interaction between its effector protein and host cells,and elucidating the complex regulation of Cpx two-component systems.The mechanism has an extremely important role and will lay the foundation for theoretical research on the pathogenesis of A.veronii in later studies.
