Background:Oral squamous cell carcinoma(OSCC),which accounts for 95%of all oral and pharyngeal cancers,is the sixth most common cancer in the world,with about 300000 new cases each year.Surgical treatment is still the main treatment,and in advanced cases,radiotherapy or chemotherapy can be used at the same time.Although these methods have made some progress in the clinical treatment of oral squamous cell carcinoma,the overall survival rate is still generally low,about 50<60%.This is mainly because in the initial clinical survey,as many as 50%of the cases had lymph node metastasis,of which nearly 50%had delayed diagnosis.Therefore,(OSCC)is still a challenging public health problem in the world,but the molecular mechanism of regulating the occurrence of oral squamous cell carcinoma is not clear.The purpose of this study was to screen the key candidate genes for export cavity squamous cell carcinoma(OSCC)by gene chip and bioinformatics method,and to explore the molecular mechanism of the occurrence and development of oral squamous cell carcinoma(OSCC).Methods:the general situation of gene expression GSE31056,GSE2280,GSE37991 and cancer genome map(TCGA)head and neck cancer was downloaded from the database of gene expression synthesis(GEO)and TCGA.R software and biological analysis kit were used to compare and identify the differentially expressed gene(DEGs).Between OSCC and normal controls.Then the gene ontology(GO)enrichment analysis of common DEGs was carried out,and the(IPA),protein-protein interaction(PPI)network analysis were analyzed by original way.The second part uses GeneBank,STRING and DAVID database to further analyze the candidate gene CXCL13 and SPP1 gene,explore the molecular mechanism of CXCL13 and SPP1 gene in the development of oral squamous cell carcinoma.Results:A total of more than 15724 differentially expressed genes were found in the chip data GSE31056.The expression of the differentially expressed genes was up-regulated and the down-regulated expression was greater than or equal to 1 and less than or equal to-1.Among them,1340 were up-regulated and 1213 were down-regulated,of which 455 were up-regulated and 1213 were down-regulated,of which 455 were up-regulated and 1213 were down-regulated,of which 841 were up-regulated and 675 were down-regulated,of which more than 2191 were up-regulated and 675 were down-regulated by chip data GSE37991.The analysis of key genes SPP1 and CXCL13 showed that SPP1 was mainly involved in extracellular matrix,cytokine activity,inflammatory reaction,osteoblast differentiation,cell adhesion and other molecular mechanisms and biological functions,involved in ECM receptor cooperation,Toll-like receptor signaling pathway,PI3K-Akt signaling pathway and cell adhesion.CXCL13 was involved in chronic inflammatory reaction,positive regulation of integrin activation and T cell chemotaxis.Endothelial cells chemotactic fibroblasts growth factor,GTPase activity activation,integrin mediated positive regulation of cell adhesion,lymph node development,cell proliferation regulation and other molecular mechanisms and biological functions,at the same time involved in cytokine receptor interaction,chemokine signaling pathway.The interaction network between CXCL13 and SPP1 protein and protein was found.Moreover,SPP1 has a certain degree of correlation in more than ten types of diseases,such as squamous cell carcinoma,inflammation,diabetes mellitus and so on.The transcription factors and related non-coding RNA are shown below,while CXCL13 is also closely related to inflammatory reaction,lymphoma,autoimmune diseases.Conclusion:Gene chip and bioinformatics method were used to explore the possible molecular mechanism of oral squamous cell carcinoma and the screening of potential molecular markers.Through bioinformatics mining of oral squamous cell carcinoma related genes,it was found that MMP family,COL11A1,CA9,SPP1 and CXCL13 may play an important role in the occurrence and development of oral squamous cell carcinoma.At the same time,some potential molecular markers related to oral squamous cell carcinoma,including SPP1 and CXCL13,were screened out and their structure and function were studied by bioinformatics.Through the analysis of gene chip and bioinformatics related genes and the screening of molecular markers,this paper provides some new ideas for the study of the pathogenesis and diagnosis and treatment of oral squamous cell carcinoma.
