Pseudomonas aeruginosa is one of the common opportunistic pathogens in clinic,which can cause high mortality in livestock,poultry and human beings.Pseudomonas aeruginosa is inherently resistant to antibiotics,and with the use of antibiotics,the increase of resistance is achieved.While strengthening the monitoring of resistance and using antibiotics accurately,phage therapy is an ideal option for reducing resistance and decreasing resistance.Bacteriophage specific infection of bacteria,there are more kinds and numbers than bacteria,and by mutation to produce more selective phages,which solves the limitation of the limited variety of antibiotics?Bacteriophages replicate in bacteria in hundreds of numbers at a time,far exceeding the efficiency of bacterial biosis.A small number of bacteriophages can cause a large number of host bacteria to lyse and die in a short period of time,and with the growth and decline of host bacteria,they are metabolized by animal organisms,thus solving the problem of antibiotic residues.In this research Pseudomonas aeruginosa and its specific lytic phages from different sources were isolated and identified.Through the epidemiological analysis of bacterial resistance,the biological properties of bacteriophage,genomics and pharmacology,the combined antibacterial action of bacteriophage and antibiotics was explored.It has been used to prevent and control rotten eggs of meat breeding chickens,and reliable results have been obtained.1.The main serotypes of 164 strains of Pseudomonas aeruginosa from different sources in shandong province were type G?27.16%?,type I?22.84%?,type B?10.49%?,and the other serotypes were more dispersed.There was no significant correlation between the flagellum type and the source of PA.The serotypes of mink,chicken and human strains were not correlated with flagellum type,while the serotypes of duck source F were correlated with flagellum type A.2.The antibiotic resistance level of Pseudomonas aeruginosa was high,and the rate of multiple antibiotic resistance was 53.3%.It is relatively sensitive to carbapenem meropenem,but has high resistance to fluoroquinolones ciprofloxacin and ofloxacin,cephalosporins cefotaxime and ceftazidine,and aminoglycosides gentamicin.The most common multiple antibiotic resistance phenotype is CIP-CTX-GEN-CAZ-OFX.3.132 bacteriophages were isolated,and the cleavage spectrum of 142 strains of Pseudomonas aeruginosa was relatively wide,and the cleavage rate was 10-52%.A broad-spectrum bacteriophage was selected for biological characterization.The incubation period was 30min,the outbreak period was about 60min,the titer was1.4×1013pfu·mL-1,the stability was good,there was no antibiotic resistant gene,toxin gene and integrase,and it had potential application value in the prevention and treatment of Pseudomonas aeruginosa infection.4.The bacteriophage pharmacology technology system and research method were established.The sensitivity of bacteriophage to antibiotics was enhanced after bacteriophage treatment.It provides a feasible way for the combination of high resistance and multi resistance strains.5.The analysis of metagenome 16SrDNA and the regression test of the isolated strains were carried out on the rotten eggs from the hatchery of meat breeding chicken.When bacteriophage coated the egg shell,the rate of rotten eggs caused by pseudomonas aeruginosa was reduced by 30%.And phage does not affect the development of breeding eggs,the safety is higher.It can provide the experimental basis for phage therapy during hatching of breeding eggs.
