In recent years,with the overdose of antibiotics,the phenomenon of bacterial resistance has become more and more serious,and the development rate of new antibiotics is much slower than the emergence of bacterial resistance.Therefore,it is necessary to find new strategies for resistant bacteria.As a virus that infects bacteria,bacteriophage has the advantages of large number and strong specificity.It is expected to replace antibiotics as an effective means to eliminate environmental pathogens.In this study,13 samples(6 sediment samples,6 water samples and 1 stalactite surface sample)from Kaiyang County,Naidong District and Panjin was collected to isolate bacteria and phages.Antibiotic resistance of host bacteria and the biological characteristics of phage was studied.At the same time,whole genome sequencing,gene function annotation and evolution analysis was performed on phage KL01.Finally,we isolated 39 bacteria and a phage KL01 with Klebsiella quasipneumoniae as the host.The host bacteria,Klebsiella quasipneumoniae,was a multi drug resistant bacteria.KL01 belongs to the family of Podoviridae;the optimal multiplicity of infection is 4;it can quickly lyse the host;it can only use Klebsiella quasipneumoniae as a host,with strong specificity.The results of genome analysis showed that the total genome size of KL01 is 45.854 kbp and the GC content is 42.5%;there were 40 ORFs in KL01,22 of which had known functions,18 of which were hypothetical proteins;it does not carry antibiotic resistance and toxicity genes;KL01 is closely related to SP6-like in Podoviridae.In this study,Klebsiella quasipneumoniae,the host bacterium,is pathogenic and can cause pyogenic liver abscess,but it is resistant to some antibiotics.Bacteriophage KL01,because of its fast cleavage speed,strong specificity and no toxic gene and resistance gene,has less impact on the environment and is safer when it is applied to the pollution control of Klebsiella quasipneumoniae in the environment.
