Various studies at home and abroad have shown that flavonoids have antiviral activity against various viruses,such as herpes virus(herpes virus,HSV),dengue virus(dengue fever virus,DENV),Zika virus(Zika virus,ZIKV),etc.Research in the field,especially against Marek's disease,is rarely reported.By reversing the virulence of Marek's disease virus(Marek's disease virus,MDV),the virulence has been continuously purified,and there have been sporadic reports of MDV incidence in chickens after immunization.The MDV vaccine currently used can prevent tumorigenesis and immunosuppression,but the virus can still replicate and disperse in the body.Therefore,the screening and research of MDV antiviral drugs,especially Chinese patent medicines,are of great significance for the prevention and control of Marek's disease virus transmission in clinic.1.The effect of flavonoids Chrysin and Genistein on the replication of Marek's disease virus in CEF cells.Previous reports have shown that the flavonoids chrysin and genistein have antioxidant,anti-tumor,anti-viral,antibacterial,anti-allergic and other effects.In order to explore the impact of these two on the replication of Marek's disease virus,experiments were designed from the following aspects:whether the drug is toxic to CEF cells and its maximum safe concentration;whether the drug has an inhibitory effect in the entire process of virus-infected cells;whether the drug Before the virus enters the cell,the process of the virus adsorbing the cell,and the virus adsorbing the cell has an inhibitory effect;whether the drug can directly affect the infectivity of the virus or even directly kill the virus.The above experiments used Real-time PCR,Western Blot and plaque count to analyze the replication of Marek virus in CEF cells.The results show that genistein can better inhibit the replication of Marek virus in CEF cells,and the inhibition effect is time and dose dependent.The results of the plaque count test show that the virus inhibition rate is 87.5%,and it can also directly affect the virus infection Sex.In addition,the expression of MDV-infected CEF cytokines was tested,and the results showed that compared with the non-genistein-treated group,genistein-induced virus-infected cells were significantly IRF-7,and IFN-? expression was also significantly increased.The two have a positive correlation.This indicates that genistein may inhibit the replication of MDV in CEF cells by inducing cells to produce IFN-?.In addition,genistein treatment significantly increased the expression of inflammatory factors IL-6 and IL-1?,suggesting that the inflammatory response of cells is also related to MDV infection.Compared with genistein,chrysin has almost no inhibitory effect on MDV replication,and even promotes MDV replication in CEF.2.Effects of baicalin(Baicalin,BA),epicatechin gallate((-)-Epicatechin gallate(ECG)and naringenin((±)-Naringenin)on the replication of Marek's disease virus in CEF cells.First,the toxicity of different drugs on CEF cells was tested,and then whether the drugs had inhibitory effect on the whole process of virus infection cells,and specifically in which stage of virus replication.Also tested whether these three drugs can directly affect the infectivity of MDV.In addition,the cytokine changes in virus-infected CEF cells were also analyzed.The experimental results show that baicalin and epicatechin gallate have a strong inhibitory effect on the replication of MDV,the inhibition rate is 90%and 91.6%,while the effect of naringenin is general,the inhibition rate is 41.6%.Cytokine test results showed that although the expression of IRF-7 was significantly reduced in the BA and ECG-treated cell groups,the expression of the antiviral factor IFN-? had no effect.This result indicates that the increased expression of IFN-?caused by MDV infection may be affected by other transcription Factor adjustment.In addition,after treatment with BA in MDV-infected CEF cells,the expression of pro-inflammatory factors IL-1? and IL-6 decreased slightly,but the difference was not significant,which may be related to the different antiviral mechanisms of BA in different cells.The expression of IL-6 in the ECG treatment group was significantly reduced,indicating that ECG can inhibit the expression of inflammatory factors3.The effect of genistein,baicalin and epicatechin gallate on the replication of Marek's disease virus in CEF cells.Compared with single flavor Chinese medicine,compound Chinese medicine may have better antiviral effect.The single flavonoids are effective in inhibiting the replication of MDV by baicalin,epicatechin gallate and genistein.They are divided into baicalin and epicatechin gallate,baicalin and genistein.These four groups are compatible with epicatechin gallate and genistein,baicalin,epicatechin gallate and genistein.First,the toxicity of these four groups of drugs to CEF cells was tested,and then whether the drugs had an inhibitory effect on the whole process of virus infection of cells was investigated.The results show that the inhibition efficiency of these groups of compound drugs is not as high as that of single drugs.
